理气补血汤调控TGF-β1/Smad通路延缓失神经骨骼肌萎缩的机制研究

摘　　要：目的探讨理气补血汤对失神经骨骼肌萎缩的作用及机制。方法将54只SD大鼠随机分为假手术组、模型组、理气补血汤组,每组18只,雌雄各半,切断右侧胫神经制作腓肠肌萎缩模型。分别在干预7、28、56 d后检测各组腓肠肌湿重比;采用HE染色和Masson染色测定肌纤维横截面积和直径、胶原纤维容积;实时荧光定量PCR法检测各组腓肠肌TGF-β1、Smad2、Smad3、MyoD1、MuRF1 mRNA表达水平;免疫荧光检测肌球蛋白的表达。结果干预7、28、56 d后,模型组和理气补血汤组腓肠肌湿重比低于同期假手术组,差异有统计学意义(P<0.05)。干预7、28、56 d后,理气补血汤组腓肠肌湿重比高于同期模型组,差异有统计学意义(P<0.05)。干预7、28、56 d后,模型组和理气补血汤组肌纤维横截面积及直径低于同期假手术组,差异有统计学意义(P<0.05);干预7、28、56 d后,理气补血汤组肌纤维横截面积及肌纤维直径高于同期模型组,差异有统计学意义(P<0.05)。干预7、28、56 d后,模型组和理气补血汤组腓肠肌胶原容积分数高于同期假手术组,差异有统计学意义(P<0.05);干预7、28、56 d后,理气补血汤组腓肠肌胶原容积分数低于同期模型组,差异有统计学意义(P<0.05)。干预7 d后,理气补血汤组与假手术组Smad2表达比较,差异无统计学意义(P>0.05);干预7、28、56 d后,模型组和理气补血汤组TGF-β1、Smad2、Smad3、MyoD1 mRNA表达高于同期假手术组,差异有统计学意义(P<0.05)。干预7、28、56 d后,理气补血汤组TGF-β1、Smad2、Smad3 mRNA表达低于同期模型组,差异有统计学意义(P<0.05)。干预7、28、56 d后,理气补血汤组MyoD1 mRNA表达高于同期模型组(P<0.05)。干预7、28、56 d后,模型组和理气补血汤组MURF1 mRNA表达量高于同期假手术组,差异有统计学意义(P<0.05);干预7、28、56 d后,理气补血汤组MURF1 mRNA表达量低于同期模型组,差异有统计学意义(P<0.05)。干预7、Objective To explore the effect and mechanism of Liqi Buxue Decoction on denervated skeletal muscle atrophy.Methods A total of 54 SD rats were randomly divided into the sham operation group(n=18),the model group(n=18)and the Liqi Buxue Decoction group(n=18).The gastrocnemius muscle atrophy model was made by cutting off the right tibial nerve.The wet weight ratio of gastrocnemius muscle was measured after 7,2856 days,the cross-sectional area and diameter of muscle fibers and the volume of collagen fibers were measured by HE staining and Masson staining,the expression of TGF-β1,Smad2,Smad3,MyoD1 and MuRF1 mRNA in gastrocnemius muscle was detected by quantitative real-time PCR,and the expression of myosin was detected by immunofluorescence.Results After 7,28,56 days of the intervention,frum muscle wet weight ratio of the model group and the Liqi Buxue Decoction group was lower than that of the sham operation group in the same period,the difference was statistically significant(P<0.05).After 7,28,56 days of the intervention,frum muscle wet weight ratio of the the Liqi Buxue Decoction group was higher than that of the model group in the same period,the difference was statistically significant(P<0.05).After 7,28,56 days of the intervention,muscle fiber cross-sectional area and diameter of the model group and the Liqi Buxue Decoction group were lower than those of the sham operation group in the same period,the differences were statistically significant(P<0.05).After 7,28,56 days of the intervention,muscle fiber cross-sectional area and diameter of the Liqi Buxue Decoction group were higher than those of the model group in the same period,the differences were statistically significant(P<0.05).After 7,28,56 days of the intervention,collagen volume fraction of the gastrocnemius muscle of the model group and the Liqi Buxue Decoction group was lower than that of the sham operation group in the same period,the difference was statistically significant(P<0.05).After 7,28,56 days of the intervention,collagen volume fraction of
