淫羊藿苷对肾癌OS-RC-2细胞增殖、迁移、凋亡及PTEN/AKT通路的影响

摘　　要：目的:探讨淫羊藿苷对肾癌OS-RC-2细胞增殖、迁移、凋亡的影响,以及对磷酸酶、张力蛋白同源物(PTEN)/蛋白激酶B(AKT)通路的影响。方法:设OS-RC-2细胞组、顺铂组(40μg/ml)和淫羊藿苷低、高浓度组(20、40μg/ml)。培养72 h后,测定各组肾癌OS-RC-2细胞存活率、迁移能力和凋亡率,PTEN、AKT的mRNA、蛋白表达水平。结果:与OS-RC-2细胞组比较,顺铂组和淫羊藿苷低、高浓度组细胞存活率降低,迁移距离缩短,PTEN、AKT的mRNA、蛋白表达水平降低,凋亡率升高,差异均有统计学意义(P<0.05)。与顺铂组比较,淫羊藿苷低浓度组细胞存活率升高,迁移距离延长,PTEN、AKT的mRNA、蛋白表达水平升高,凋亡率降低;淫羊藿苷高浓度组细胞存活率降低,迁移距离缩短,PTEN、AKT的mRNA、蛋白表达水平降低,凋亡率升高,差异均有统计学意义(P<0.05)。与淫羊藿苷低浓度组比较,淫羊藿苷高浓度组细胞存活率降低,迁移距离缩短,PTEN、AKT的mRNA、蛋白表达水平降低,凋亡率升高,差异均有统计学意义(P<0.05)。结论:淫羊藿苷能够抑制肾癌OS-RC-2细胞增殖和迁移,促进肾癌OS-RC-2细胞凋亡,其机制可能与淫羊藿苷降低肾癌OS-RC-2细胞PTEN、AKT的mRNA、蛋白表达,进而抑制PTEN/AKT通路的激活有关。OBJECTIVE:To probe into the effects of icariin on proliferation,migration,apoptosis and phosphatase and tensin homolog deleted on chromosome ten(PTEN)/protein kinase B(AKT)pathway of renal cancer OS-RC-2 cells.METHODS:OS-RC-2 cell group,cisplatin group(40μg/ml),icariin groups with different concentrations(20μg/ml,40μg/ml)were set up.After 72 h of culture,the survival rate,migration ability,apoptosis rate,and the mRNA and protein expression levels of PTEN and AKT were measured in each group of renal cancer OS-RC-2 cells.RESULTS:Compared with the OS-RC-2 cell group,the cell survival rate,migration distance,the mRNA and protein expression levels of PTEN and AKT decreased,and the apoptotic rate increased in the cisplatin group,icariin low-concentration group and icariin high-concentration group,with statistically significant differences(P<0.05).Compared with the cisplatin group,the cell survival rate,migration distance,the mRNA and protein expression levels of PTEN and AKT increased and the apoptotic rate decreased in the icariin low-concentration group;the cell survival rate,migration distance,the mRNA and protein expression levels of PTEN and AKT decreased and the apoptotic rate increased in the icariin high-concentration group,with statistically significant differences(P<0.05).Compared with the icariin low-concentration group,the cell survival rate,migration distance,the mRNA and protein expression levels of PTEN and AKT decreased and the apoptotic rate increased in the icariin high-concentration group,with statistically significant differences(P<0.05).CONCLUSIONS:Icariin can inhibit the proliferation and migration of renal cancer OS-RC-2 cells,and promote the apoptosis.The mechanism may be related to the reduction of the mRNA and protein expression levels of PTEN and AKT in renal cancer OS-RC-2 cells by icariin,which in turn inhibits the activation of PTEN/AKT pathway.
