摘  要:为开发新型微生物来源的新型抑制植物病毒的生防菌剂,本研究利用来源于青稞酒糟的菌株发酵液进行抑制马铃薯Y病毒(Potato Virus Y,PVY)活性的测定,并进行活性菌株的鉴定。Real-Time PCR实验结果表明:菌株JZ1-4-2、JZ2-1-13和JZ1-1-2对PVY具有较高的抑制活性,抑制率分别为99.95%、99.61%和99.07%;菌株JZ2-4-2和JZ3-1-12对PVY具有中等的抑制活性,抑制率分别为52.70%和57.37%。对3株活性菌株JZ1-4-2、JZ2-1-13和JZ1-1-2进行分子生物学鉴定,结果显示,菌株JZ1-4-2为枯草芽孢杆菌(Bacillus subtilis subsp.subtilis)、菌株JZ2-1-13为解淀粉芽孢杆菌(Bacillus amyloliquefaciens)、菌株JZ1-1-2为巴基斯坦芽孢杆菌(Lysinibacillus pakistanensis)。综上所述,从青稞酒糟中分离到的菌株JZ1-4-2、JZ2-1-13和JZ1-1-2具有作为PVY生防菌剂开发的潜力。本研究结果为PVY生防菌剂的研发提供理论依据和技术支持。In order to develop a new type of microbial-derived biocontrol agent that inhibits plant viruses,this study used the fermentation broth of the strain derived from highland barley distiller’s grain to determine the inhibitory activity against Potato Virus Y(PVY)and identify the active strains.Real-Time PCR experiment results showed that:strains JZ1-4-2,JZ2-1-13 and JZ1-1-2 had high inhibitory activity against PVY,with inhibition rates 99.95%,99.61%and 99.07%,respectively;strains JZ2-4-2 and JZ3-1-12 had moderate inhibitory activity against PVY,with inhibition rates 52.70%and 57.37%,respectively.The molecular biological identification of three active strains JZ1-4-2,JZ2-1-13 and JZ1-1-2 showed that the strain JZ1-4-2 is Bacillus subtilis subsp.subtilis.,strain JZ2-1-13 is Bacillus amyloliquefaciens,and strain JZ1-1-2 is Lysinibacillus pakistanensis.In summary,the strains JZ1-4-2,JZ2-1-13 and JZ1-1-2 isolated from highland barley distiller’s grain have the potential to be developed as PVY biocontrol agents.The results of this study provide theoretical basis and technical support for the research and development of PVY biocontrol agents.
