摘 要:为明确培养时间、培养基pH值、蔗糖、硼酸、PEG-4000对百香果花粉离体萌发的影响,筛选出花粉体外萌发的适宜条件、优化液体培养基组分,本研究以‘台农一号’百香果花粉为材料,采用液体培养基离体培养的方法,开展单因素及正交试验,并用‘台农一号’及‘黄果’百香果的花粉验证了最适宜培养基的萌发率。结果显示:培养时间、培养基pH值、蔗糖、硼酸、PEG-4000对花粉萌发率及花粉管生长均有显著的影响。培养时间为4 h,培养基pH值为5.5时萌发率最高,蔗糖、硼酸、PEG-4000为百香果花粉离体萌发的必需物质,适当的浓度可促进花粉萌发及花粉管的生长,但浓度过高起抑制作用,因素间存在明显的互作效应。百香果花粉离体萌发的最佳培养基为:150 g/L蔗糖+150 g/L PEG-4000+25 mg/L H_(3)BO_(3)+300 mg/L Ca(NO_(3))_(2)·4H_(2)O+200 mg/L Mg SO_(4)·7H_(2)O+100 mg/L KNO_(3),‘台农一号’百香果及‘黄果’百香果花粉在该培养基中的离体萌发率分别为95.99%及92.35%。本研究筛选到的适宜百香果花粉体外萌发的培养条件及萌发率高的液体培养基,为生产及杂交育种中的人工授粉及百香果孢粉学研究提供参考。In order to clarify the influences of culture time,medium pH value,sucrose,boric acid and PEG-4000 on the in vitro germination of passion fruit pollen,the suitable conditions for pollen germination in vitro were screened and the components of liquid medium were optimized.In this study,the pollen of"Tainong No.1"pas-sion fruit was used as material,and the single factor and orthogonal test were carried out by the method of liquid medium culture in vitro.The pollen of'Tainong No.1'and'Huangguo'passion fruit was used to verify the germina-tion rate of the most suitable medium.The results showed that the culture time,medium pH value,sucrose,boric acid and PEG-4000 have a significant influence on pollen germination rate and pollen tube growth.When the cul ture time was 4 h,the medium pH value was 5.5,the germination rate was maximum.Sucrose,boric acid and PEG-4000 were essential substances for pollen germination of passion fruit in vitro.Appropriate concentration could promote pollen germination and pollen tube growth,but too high concentration had an inhibited effect,an obvious interaction effect existed among them.The optimal medium for pollen germination of passion fruit in vitro was 150 g/L sucrose+150 g/L PEG-4000+25 mg/L H_(3)BO_(3)+300 mg/L Ca(NO_(3))_(2)·4H_(2)O+200 mg/L Mg SO_(4)·7H_(2)O+100 mg/L KNO_(3).In vitro germination rate of passion fruit pollen of'Tainong No.1'and yellow fruit passion fruit(P.edulis f.flavicarpa Deg.)in this medium were 95.99%and 92.35%,respectively.In this study,suitable cultural conditions and liquid medium with high germination rate of passion fruit pollen in vitro were selected,which provide refer-ence for artificial pollination and palynology research of passion fruit in production and cross breeding.
