摘  要:目的研究并比较小鼠洛哌丁胺便秘模型和小鼠低纤维膳食便秘模型。方法2种模型(洛哌丁胺便秘模型组或低纤维膳食便秘模型组)各选用昆明种小鼠40只,按体重随机分成4组,每组10只,其中2组动物用于小肠运动实验和检测胃肠功能相关激素水平,另2组动物用于小鼠排便试验。建模后进行小肠运动实验、排便实验及胃肠功能相关激素水平测定。结果洛哌丁胺便秘模型:小肠运动实验结果显示洛哌丁胺组(3 mg/kg·bw)与阴性对照组比较,小鼠洋红推进率降低(P<0.01);小鼠排便实验结果显示,洛哌丁胺组(6 mg/kg·bw)与阴性对照组比较,6 h排便粒数减少(P<0.01)、粪便湿重降低(P<0.05),排便时间延长(P<0.01);与阴性对照组比较,洛哌丁胺模型组小鼠血清P物质(SP)水平降低(P<0.05)、内皮素-1(ET-1)水平降低(P<0.05)、胃动素(MTL)水平降低(P<0.01)、胃泌素(GAS)水平降低(P<0.05)和乙酰胆碱酯酶(AchE)水平降低(P<0.05)。低纤维膳食便秘模型:LFD组与普通饲料对照组比较,小鼠小肠推进率差异无统计学意义(P>0.05);小鼠排便实验结果显示,与普通饲料对照组比较,LFD组小鼠6 h排便粒数减少(P<0.01)、粪便湿重降低(P<0.01)、排便时间延长(P<0.01),与普通饲料对照组比较,LFD组小鼠血清SP水平降低(P<0.01)、ET-1水平降低(P<0.05)和MTL水平降低(P<0.05);小鼠血清SS水平明显升高(P<0.01)。结论洛派丁胺法可成功复制小肠运动实验和小鼠排便实验便秘模型,低纤维膳食可成功复制小鼠排便试验小鼠便秘模型,但小肠运动实验便秘模型不成立。2种小鼠便秘模型引起胃肠激素改变的种类不完全相同,这可能与2种便秘模型引发便秘的机制不同有关。Objective To study and compare loperamide constipation model with low fiber dietary constipation model in mice.Methods Forty mice in each of the two models(loperamide constipation model group or low fiber dietary constipation model group)were randomly divided into four groups according to mice body weight,with 10 mice in each group.Two groups of mice were used for intestinal motility test and gastrointestinal function related hormone detection,and the other two groups of mice were used for defecation test.After the models were established,intestinal motility test,defecation test and gastrointestinal function related hormone levels were measured.Results In the loperamide constipation model,the intestinal motility test result showed that the carmine gastrointestinal transit ratio of mice in the loperamide group(3 mg/kg·bw)was lower than that in the negative control group(P<0.01).The result of defecation test in mice showed that compared with the negative control group,the number of defecation particles was decreased(P<0.01),the wet weight of feces was decreased(P<0.05),and the defecation time was prolonged(P<0.01)in loperamide group.Compared with the negative control group,the serum SP(P<0.05),ET-1(P<0.05),MTL(P<0.01),GAS(P<0.05)and AchE(P<0.05)levels in loperamide model control group were decreased.In the low fiber dietary constipation model:gastrointestinal transit ratio of mice in LFD group had no statistically significant difference with normal diet control group(P>0.05);The result of defecation experiment in mice showed that compared with normal diet control group,the number of defecation feces in LFD group was decreased(P<0.01),the wet weight of feces was decreased(P<0.01),and the defecation time was prolonged(P<0.01).Compared with normal diet control group,the serum SP,ET-1,and MTL levels in LFD group were decreased(P<0.01);The serum SS level of mice was significantly increased(P<0.01).Conclusion Loperamide can be used to replicate the constipation model of mice in small bowel movement experiment and defeca
