摘 要:目的:初步研究肾脏雄激素调节蛋白(KAP)在单侧输尿管梗阻(UUO)小鼠肾纤维化进展中的作用。方法:我们首先在基因表达综合数据库(GEO)下载GSE62732小鼠肾纤维化模型芯片,利用Limma函数包筛选出差异基因;然后通过手术结扎C57小鼠单侧输尿管构建UUO小鼠肾纤维化模型;进一步通过免疫组化和蛋白免疫印迹实验检测上述模型中KAP的表达情况。结果:在GSE62732小鼠肾纤维化模型中筛选获得了差异基因KAP;成功构建了本实验的UUO肾间质纤维化小鼠模型;免疫组化及蛋白免疫印迹实验均发现本实验UUO小鼠肾纤维化模型中KAP蛋白表达水平明显升高。结论:KAP在UUO小鼠肾纤维化模型中的表达水平升高。Objective:To investigate the role of kidney androgen-regulated protein(KAP)in the progression of renal fibrosis in mice with unilateral ureteral obstruction(UUO).Methods:GSE62732mouse renal fibrosis model microarray was downloaded from the Gene Expression Omnibus(GEO)database,and the differential genes were screened out by Limma function packet.Then,the renal fibrosis model of UUO mice was established by surgical ligation of unilateral ureter of C57mice.The expression of KAP in the above models was further detected by im-munohistochemistry and western blot.Results:The gene KAP was obtained from GSE62732mouse renal fibrosis model.A mouse model of UUO renal interstitial fibrosis was successfully established.Immunohistochemistry and Western blot assay showed that the expression level of KAP in UUO mouse renal fibrosis model was significantly increased.Conclusion:The expression level of KAP in UUO mouse model of renal fibrosis is increased.
