摘  要:目的了解济南市市售肉鸡中空肠弯曲菌的污染情况、分离株的分子分型特征及其耐药性和毒力基因携带情况。方法采用滤膜法于2015—2018年对济南市农贸市场售卖的98份现杀活鸡整禽中的空肠弯曲菌进行分离培养;利用脉冲场凝胶电泳和多位点序列分型对分离株进行分子分型;用商品化药敏板测试分离株对11种八类药物的敏感性;通过PCR对5种毒力相关基因进行筛查分析。结果济南市市售肉鸡中空肠弯曲菌的检出率为26.5%(26/98);PFGE分型的结果显示,济南市肉鸡来源26株空肠弯曲菌共有16种带型,带型的相似度在53.9%~100%之间;MLST分型的结果显示,26株空肠弯曲菌可分为19种ST型,其中新发现型别占比为47.4%(9/19)。药敏测试的结果显示,空肠弯曲菌对治疗首选药物喹诺酮类抗生素环丙沙星和萘啶酸的耐药率高达92.3%和100%;分离株五种毒力基因:cad F、cdt B、iam A、fla A和virBII的携带率为分别为100%、100%、92.3%、69.2%和19.2%。结论济南市市售肉鸡中存在一定程度的空肠弯曲菌污染,分离株存在遗传多样性,耐药情况比较严重,且携带有毒力基因的菌株的比例较高,因此应加强空肠弯曲菌感染监测。Objective To investigate the contamination of Campylobacter jejuni,molecular typing characteristics,drug resistance and virulence gene carrying rate of Campylobacter jejuni isolates from broilers for sale in Jinan city.Methods Campylobacter jejuni was isolated and cultured from 98 live chickens sold in Jinan farmers ’ market in 15-18 years by filtration membrane method.Molecular typing of the isolates was performed by pulsed field gel electrophoresis and multilocus sequence typing.The sensitivity of the isolates to eight classes of 11 drugs was tested by commercial drug sensitive plate.Five virulence related genes were screened and analyzed by PCR.Results The results of PFGE typing showed that there were16 bands of Campylobacter jejuni strains from 26 broilers in Jinan,and the similarity of the bands ranged from 53.9% to100%.The results of MLST typing showed that the 26 Campylobacter jejuni strains could be classified into 19 ST types,among which the newly discovered type accounted for 47.4%(9/19).The drug susceptibility test results showed that the resistance rates of Campylobacter jejuni to the preferred quinolone antibiotics ciprofloxacin and nalidixic acid were as high as92.3% and 100%.The carrying rate of five virulence genes Cad F,Iam A,Cdt B,Vir BII,Fla A were 100%,92.3%,100%,19.2% and 69.2%,respectively.Conclusion There is a certain degree of Campylobacter jejuni contamination in broilers sold in Jinan.The isolates have genetic diversity and serious drug resistance,and the proportion of strains carrying virulence genes is relatively high.Therefore,it is necessary to strengthen the contamination and infection monitoring of Campylobacter jejuni and formulate appropriate drug use strategies.
