摘 要:目的研究黄芩黄酮Ⅰ对结直肠癌细胞迁移、侵袭和上皮-间质转化(Epithelial-mesenchymal transition,EMT)的影响。方法取对数生长期结直肠癌LoVo细胞,将其随机分为对照组和实验组,分别用DMEM培养液和含80μmol·L^(-1)黄芩黄酮Ⅰ的DMEM培养液处理。用划痕实验检测LoVo细胞的迁移能力,用Transwell法检测LoVo细胞的侵袭能力,用蛋白质印迹法检测LoVo细胞中上皮型钙黏蛋白(E-cadherin)、神经型钙黏蛋白(N-cadherin)和波形蛋白(Vimentin)的表达,用荧光定量聚合酶链反应(PCR)法检测LoVo细胞miR-378的相对表达。结果对照组与实验组细胞迁移率分别为(61.79±1.24)%和(46.18±1.41)%;细胞侵袭数分别为(79.96±1.55)和(58.41±0.98)个;E-cadherin蛋白表达量分别为1.01±0.03和2.60±0.07;N-cadherin蛋白的表达量为0.99±0.02和0.45±0.03;Vimentin蛋白的表达量为0.99±0.02和0.52±0.04;miR-378相对表达量分别为0.43±0.03和0.85±0.02,差异均有统计学意义(均P<0.05)。结论黄芩黄酮Ⅰ可显著抑制结直肠癌细胞的迁移、侵袭和EMT,其作用机制可能与上调miR-378表达有关。Objective To elaborate the effects of scutellaria baicalensis flavone Ⅰ on migration, invasion, and epithelial-mesenchymal transition(EMT) of colorectal cancer cells. Methods Colorectal cancer cells LoVo in logarithmic growth phase were randomly divided into control group and experimental group, which were treated with DMEM medium and DMEM medium containing 80 μmol·L;scutellaria baicalensis flavone Ⅰ, respectively. The migration and invasion of LoVo cells were detected by wound healing assay and Transwell assay. The expression of E-cadherin, N-cadherin and Vimentin in LoVo cells were detected by Western blot assay. The relative expression of miR-378 in LoVo cells was detected by fluorescence quantitative polymerase chain reaction. Results The cell migration rates in control group and experimental group were(61.79±1.24) % and(46.18±1.41) %, the number of cell invasion were 79.96±1.55 and 58.41±0.98, the expression of E-cadherin protein were 1.01±0.03 and 2.60±0.07, the expression level of N-cadherin protein were 0.99±0.02 and 0.45±0.03, the expression levels of Vimentin were 0.99±0.02 and 0.52±0.04, the relative expression levels of miR-378 were 0. 43 ± 0. 03 and 0. 85 ± 0. 02, the differences were all statistically significant(all P< 0. 05).Conclusion Scutellaria baicalensis flavone Ⅰ can significantly inhibit the migration,invasion and EMT of colorectal cancer cells, and the mechanism may be related to the upregulation of miR-378 expression.
