RNA干扰AC-cathB-1基因对广州管圆线虫Ⅰ期幼虫发育的影响

摘  要:明确广州管圆线虫组织蛋白酶B样基因1(AC-cathB-1)功能,为揭示广州管圆线虫致病机制及生物防治提供理论依据。利用RNA干扰技术抑制广州管圆线虫Ⅰ期幼虫AC-cathB-1基因的表达,测定该基因被干扰后对福寿螺的感染率、死亡率、虫体活力、发育成Ⅲ期幼虫的数量及虫体长度的影响。real-time PCR检测结果显示,AC-cathB-1干扰组基因相对表达量明显下调,AC-cathB-1干扰组与生理盐水对照组的差异显著(P<0.05);与Rubisco干扰组、生理盐水对照组相比,AC-cathB-1干扰组福寿螺的感染率分别降低了10%和16.67%,死亡率分别降低了16.67%和13.34%,虫体活力减弱;AC-cathB-1干扰组的Ⅲ期幼虫的数量分别减少了11.4条和15.1条,发育率降低了34.97%和41.60%。与生理盐水对照组均差异显著(P<0.05)。AC-cathB-1干扰组的Ⅲ期幼虫体长有所减短,但与生理盐水对照组差异不显著(P>0.05)。结果表明,RNA干扰广州管圆线虫Ⅰ期幼虫AC-cathB-1基因可一定程度上抑制Ⅰ期幼虫在中间宿主福寿螺体内发育成Ⅲ期幼虫。证明了AC-cathB-1基因在寄生虫入侵宿主和宿主的相互作用等方面发挥着重要的作用。By studying the function of the cathepsin B-like 1 cysteine protease gene(AC-cathB-1)of A.cantonensis,we provided a theoretical basis for revealing the pathogenic mechanism and biological control of A.cantonensis.RNA interference was used to investigate the effect of silencing of AC-cathB-1 gene expression in stage-Ⅰlarvae of A.cantonensis on infection rate,mortality of Pomacea canaliculata,viability,the number of stage-Ⅲlarvae and length.The real-time PCR results showed that the relative expression of genes in the AC-cathB-1 interference group was significantly downregulated,and the difference between the AC-cathB-1 interference group and the control group was statistically significant(P>0.05).Compared with Rubisco interference group and saline control group,the infection rate of Pomacea canaliculata of the AC-cathB-1 interference group decreased by 10%and 16.67%,and mortality rates decreased by 16.67%and 13.34%,respectively.The viability of stage-Ⅲlarvae was reduced.The number of stage-Ⅲlarvae in the AC-cathB-1 interference group was reduced by 11.4 and 15.1,and the rate decreased by 34.97%and 41.60%,respectively.The differences were statistically significant(P<0.05).The length of stage-Ⅲlarvae of the AC-cathB-1 interference group was reduced,but it was not statistically significant compared with the control group.The results of the study showed that RNAi with the AC-cathB-1 gene of the stage-Ⅰlarvae,to a certain extent,could inhibit the development of the stage-Ⅰlarvae into the stage-Ⅲlarvae in the intermediate host of Pomacea canaliculata.It is proved that the AC-cathB-1 gene plays an important role in the invasion of the host and the interaction of the host.
