血清7型副猪嗜血杆菌的分离鉴定及生物学特性研究

摘　　要：为了对疑似副猪嗜血杆菌感染的仔猪病例进行诊治,对病死仔猪进行了细菌分离,用16S rRNA测序、套式PCR、血清型分型PCR、基因型分型PCR等方法对分离菌株进行鉴定,并对分离菌株进行药敏试验和小鼠致病性试验。结果显示,从发病仔猪的关节液中分离得到1株革兰氏阴性菌;16S rRNA测序和特异性套式PCR结果表明,该分离菌株为副猪嗜血杆菌;血清型分型PCR结果表明,分离菌株为血清型7型;多点位序列分型ST-400;分离菌株对头孢曲松、阿莫西林/棒酸、庆大霉素、壮观霉素、多西环素、甲氧苄啶/磺胺甲恶唑、左氟沙星、环丙沙星、氟苯尼考、多黏菌素B敏感,对亚胺培南、林可霉素耐药;小鼠致病性试验结果表明,该菌株致病性和毒力较低。结果表明,在饲养条件不良或者有混合感染的条件下,血清7型副猪嗜血杆菌可能感染猪群并表现出相应的临床症状,可以根据药敏试验的结果选择头孢曲松、氟苯尼考等敏感药物对患病仔猪进行用药治疗。试验证实了血清7型副猪嗜血杆菌在临床中的感染情况,并为临床上对该病的治疗用药提供了一定参考依据。In order to diagnose and treat piglets suspected of being infected with Haemophilus parasuis,this study isolated the bacteria from dead piglets and used 16S rRNA sequencing,nested PCR,serotyping PCR,and genotyping PCR methods to analyze strain for identification,and conduct antimicrobial susceptibility test and mouse pathogenicity test on the isolated strain.The results showed that a Gram-negative bacterium was isolated from the synovial fluid of affected piglets;16S rRNA sequencing and specific nested PCR results showed that the isolated strain was Haemophilus parasuis;the serotype PCR results showed that the isolated strain is serotype 7;Multipoint sequence typing is ST-400.The results of the antimicrobial susceptibility test showed that the isolated strain was sensitive to ceftriaxone,amoxicillin/clavulanic acid,gentamicin,spectinomycin,doxycycline,trimethoprim/sulfamethoxazole,levofloxacin,ciprofloxacin,florfenicol,and polymyxin B.On the contrary,it is highly resistant to imipenem and lincomycin.The mouse pathogenicity test results showed that the strain is low in pathogenicity and virulence.The above results indicated that under poor feeding conditions or mixed infections,Haemophilus parasuis serotype 7 may infect pigs and show corresponding clinical symptoms.Pig farms can choose sensitive drugs such as ceftriaxone and florfenicol to treat sick piglets based on the results of the antimicrobial susceptibility test.This study confirmed the clinical infection of serotype 7 Haemophilus parasuis,and provided a certain reference for the clinical treatment of the disease.
