摘 要:蛋白质间的相互作用在预测靶蛋白功能、了解病毒感染宿主的机制和研发新型抗病毒药物方面发挥着重要作用。大多数蛋白质以复合物形式来实现其功能。近年来,已经研发了多种基于生物物理、生物化学或遗传学的方法来辅助蛋白质-蛋白质相互作用(PPI)的研究,丰富了PPI数据,为研究蛋白互作提供了重要依据。每种检测方法在敏感性和特异性方面都有自身的优点和局限性。论文围绕几种经典的蛋白互作研究方法,包括表面等离子体共振、免疫共沉淀、谷胱甘肽转移酶沉淀试验和酵母双杂交等,概述其原理,分析其优点、局限性及在病毒学领域的最新应用,以期为研究人员选择合适的PPI研究技术提供参考。Protein interaction plays a key role in predicting the function of the target protein,understanding the mechanism of virus infecting hosts and researching new antiviral drugs.Most proteins fulfill their phenotypic functions in the form of complexes.In recent years,a variety of methods based on biophysics,biochemistry and genetics have been developed to assist the study of protein-protein interaction(PPI),enriching PPI data and providing important information for studying protein-protein interactions.However,each detection method has its own advantages and limitations in terms of sensitivity and specificity.The thesis discussed several classic protein interaction research methods,including Surface plasmon resonance(SPR),Co-immunoprecipitation(Co-IP),Glutathione S-transferase pull-down(GST pull-down)and Yeast two-hybrid(Y2H),etc,and summarized their principles,analyzed their advantages,limitations,and their latest applications in the field of virology,in order to provide a reference for researchers to choose appropriate PPI research techniques.
