摘  要:为确定新疆南疆某羊场羊感染羊棘球蚴、细颈囊尾蚴的基因型,对新疆南疆某羊场1例棘球蚴、细颈囊尾蚴混合感染进行分子生物学鉴定。在临床诊断、剖检、镜检观察的基础上,分别采用棘球蚴COI、NDI基因和细颈囊尾蚴Cytb基因的引物进行PCR扩增、测序、核苷酸同源性分析,并构建系统进化树。结果显示,该混合感染样品棘球蚴COI、NDI基因序列与GenBank中棘球蚴Sheep(G1)株(EF367301.1)高度同源,核苷酸同源性为100%,同属于棘球蚴G1型;细颈囊尾蚴Cytb基因序列与GenBank中细颈囊尾蚴IM4株(KT258035.1)高度同源,同源性为99.5%。结果表明,该病例为羊细粒棘球蚴G1基因型和细颈囊尾蚴的混合感染。The purpose of this study was to identify the hydatid cycst and Cysticercus tenuicollis mixed in sheep of southern Xinjiang.On the basis of clinical diagnosis,the COI,NDI genes of hydatid cyst and Cytb gene of Cysticercus tenuicollis were amplified respectively by PCR with special primers.COI,NDI and Cytb genes were analyzed by bioinformatics software respectively.The phylogenetic tree was constructed by Neighbor-joining.The results showed that the COI and NDI gene sequences of hydatid cyst in the mixed infection samples are highly homologous to the sheep(G1)strain(EF367301.1)of hydatid cyst in GenBank,with 100%nucleotide homology.Both of them belonge to the G1 genotype.The Cytb gene sequence of Cysticercus tenuicollis is highly homologous with the IM4 strain(KT258035.1)and the homology was 99.5%.This study confirmed that the case was concurrently infected with hydatid cycst G1 genotype and Cysticercus tenuicollis.
