摘 要:为探讨鱼腥草挥发油对膝关节骨性关节炎大鼠血清IL-1β、IL-6和TNF-α水平的影响,选取60只健康SD大鼠,雌雄各半,分为假手术组(正常对照组)、模型组(KOA模型对照组)、阳性药物组(阳性药物对照组)、鱼腥草挥发油高中低剂量组,每组10只。采用改良Hulth法建立膝关节骨性关节炎模型,造模4周后,阳性药物组给予盐酸氨基葡萄糖片水溶液灌胃治疗,鱼腥草挥发油高、中、低剂量组分别给予由吐温-80配制成的鱼腥草挥发油制备液灌胃治疗,模型组和正常组给予同阳性药物组相同剂量的蒸馏水灌胃。连续给药6 w,1次/d,后取各组大鼠血清,ELISA法检测血清IL-1β、IL-6和TNF-α水平。取膝关节组织,镜下观察组织病理学改变。结果发现,6 w给药治疗结束后,与正常组比较,模型组和各药物治疗组大鼠血清IL-1β、IL-6和TNF-α水平均显著升高(P<0.05);与模型组比较,各药物治疗IL-1β、IL-6和TNF-α水平均有不同程度的降低。其中,阳性药物组和高剂量组炎症因子水平降低最为明显,两组间比较,差异不具有统计学意义(P>0.05);病理观察显示,正常组大鼠膝关节滑膜未见异常改变,模型组大鼠膝关节滑膜组织内可见纤维组织明显增生及大量炎症细胞浸润,高剂量组间质轻度纤维化,慢性炎症细胞浸润现象明显好转,与阳性对照组病变缓解程度相近。说明鱼腥草挥发油能够通过降低IL-1β、IL-6和TNF-α炎症因子水平,改善KOA大鼠膝关节滑膜组织的病理学改变,发挥缓解炎症的作用。In order to investigate the effect of volatile oil of Houttuynia cordata on the level of serum IL-1β,IL-6 and TNF-αin rats with knee osteoarthritis.60 healthy SD rats,including half male and half female,were divided into sham operation group(normal control group)and model group(KOA model control group);positive drug group(positive drug control group);high,medium and low dose Houttuynia volatile oil groups,with 10 rats in each group.Improved Hulth method was used for knee osteoarthritis model construction.After 4-weak modelling,the positive drug group was given glucosamine hydrochloride tablets by gavage;the high,middle and low dose groups were given the preparation of volatile oil of Houttuynia cordata prepared by Tween-80 by gavage.The model group and the normal group were given the same dose of distilled water by gavage as the positive drug group,and the drug was given for 6 weeks,once a day.After that,the serum of rats in each group was taken,and the serum was detected by ELISA IL-1β,IL-6 and TNF-α.The histopathological changes were observed under the microscope.The results show that after 6 weeks of treatment,compared with the normal group,the levels of serum in the model group and each drug treatment group were significantly lower,the levels of IL-1β,IL-6 and TNF-αincreased significantly(P<0.05).Compared with the model group,the levels of IL-1β,IL-6 and TNF-αin the treatment group decreased in different degrees;among them,inflammatory factors levels of positive medicine group and high dosage group decreased most significantly,with no statistically significant difference(P>0.05).Pathological observation showed that there was no abnormal change in the synovium of knee joint in the normal group,fibrous tissue proliferation and a large number of inflammatory cells infiltration were found in the synovium of knee joint in the model group,interstitial fibrosis was mild in the high-dose group,and chronic inflammatory cell infiltration improved significantly,which was similar to the remission degree of the po
