摘 要:为研究陆地棉杯状叶变异的分子遗传规律,以自主鉴定的陆地棉绿色杯状叶突变体‘R169’为试验材料,通过与海岛棉品种‘胜利1号’配制海陆杂交组合进行遗传分析,结果显示,杯状叶突变体为单位点质量性状突变体,符合由一对隐性基因控制的孟德尔分离规律。而后利用棉花单位点质量基因在染色体的快速定位方法对该突变体基因(cup)进行染色体定位,对12个特定样本组成的小群体分析显示,引物NAU3480、BNL1066与目的基因cup连锁。由于引物NAU3480、BNL1066均位于棉花第11染色体,据此推断cup基因位于第11染色体。而后合成位于该染色体的110对SSR引物,通过对双亲DNA的PCR扩增得到26对多态性引物。用这些引物检测F2代群体每个单株的标记基因型,然后通过软件进行遗传连锁分析,结果表明共有19对引物与基因cup连锁,它们分别是引物NAU5192、NAU3621、NAU2118、NAU1162、NAU3409、NAU2998、NAU3657、NAU1148、NAU3234、NAU2651、NAU2599、NAU2661、NAU1014、NAU3622、BNL1408、NAU4086、BNL1066、NAU3480、NAU5505。基因cup位于SSR分子标记BNL1066与NAU3480之间,遗传距离分别为9.3 cM和1.6 cM。To study the molecular genetic law of a cup leaf mutant in Gossypium hirsutum, a self-identified cotton cup leaf mutant‘R169’was used as the experimental material, the sea-land hybrid combination was conducted with the island cotton variety‘ Shengli 1’. The genetic analysis showed that the cup leaf mutant was a mono-locus quality character mutant, controlled by a pair of recessive genes and fit the Mendel pattern.Then the method of rapid mapping of the mono-locus quality character gene on chromosome was used to locate the mutant cup gene. The results by checking 12 specific samples showed that the primers NAU3480 and BNL1066 were linked to the cup gene. As the primers NAU3480 and BNL1066 were located on the 11 th chromosome of cotton, it was inferred that the cup gene was located on the 11 thchromosome. Then 110 pairs of primers on the chromosome were synthesized and 26 pairs of polymorphic primers were obtained by PCR parental DNA. These primers were used to detect the marked genotype of each individual plant in F2 population and then genetic linkage analysis was performed by software. The results showed that 19 pairs of primers were linked to the gene cup, which were NAU5192, NAU3621, NAU2118, NAU1162, NAU3409,NAU2998, NAU3657, NAU1148, NAU3234, NAU2651, NAU2599, NAU2661, NAU1014, NAU3622,BNL1408, NAU4086, BNL1066, NAU3480, NAU5505. Gene cup was located between BNL1066 and NAU3480, and the genetic distance was 9.3 c M and 1.6 c M, respectively.
