摘  要:为明确绿豆愈伤组织培养体系中最佳种子消毒方式,利用乙醇、次氯酸钠(NaClO)、氯化汞(HgCl_(2))和氯气(Cl_(2))作为消毒剂,组合搭配16种消毒方式对绿豆种子进行消毒处理。对发芽率、污染率、种皮破损率及愈伤组织诱导率等指标进行综合评价分析。结果表明,75%乙醇30 s+1.0%NaClO 10 min的消毒方式处理的绿豆种子发芽率为90.11%,该处理无污染,种皮无破损,出愈率达到92%,诱导的愈伤组织呈黄绿色块状,结构紧密且质地硬,生长速率与生长状况良好。因此,该消毒方式适用于绿豆愈伤组织培养过程中的种子消毒,且有利于绿豆子叶节的愈伤诱导。In order to determine the best disinfection method of mung bean seeds in callus culture system,16 disinfection methods were used to disinfect mung bean seeds with ethanol/NaClO/HgCl_(2) and chlorine as disinfectants.In addition,the germination rate,pollution rate,seed coat damage rate and callus induction rate were comprehensively evaluated and analyzed.The results showed that the germinating rate of mung bean seeds treated with 75%ethanol for 30 s+1.0%NaClO for 10 min was 90.11%,and there was no pollution,the seed coat was not damaged,the callus induction rate was 92%,and the callus was light yellow green,compact in structure and hard in texture.Therefore,this method was suitable for seed disinfection in the process of mung bean callus culture and is conducive to the callus induction of mung bean cotyledon node.
