摘 要:本文以胡桃科黄杞属植物黄杞的干燥叶为原料,落新妇苷为定量指标,采用微波辅助法醇提取黄杞叶,在单因素试验的基础上采用三因素三水平的Box-Behnken实验设计,优化提取工艺。通过体外自由基清除能力、还原能力和抗脂质膜氧化作用考察黄杞叶醇提物的抗氧化活性。结果表明,黄杞叶最优醇提取工艺为:乙醇浓度64%、微波温度56℃、微波时间67s。黄杞叶醇提物浓度高于0.8 mg/mL时,还原能力及ABTS+自由基清除能力明显强于落新妇苷(P<0.05),实验浓度范围内醇提物对抗脂质膜的氧化作用显著优于落新妇苷(P<0.01)。本实验表明,响应面法对黄杞叶醇提取工艺优化具有可行性,黄杞叶醇提物有一定的抗氧化活性,在生物医药领域有潜在应用前景。The dried leaves of Engelhardtia roxburghiana Wall. of the Juglans family were extracted by microwave-assisted method with alcohol. The process was evaluated according to the content of astilbin in the extracts. On the basis of single factor experiment, Box-Behnken experimental design with three factors and three levels was employed to optimize the extraction process. The antioxidant activities of the alcohol extract from the leaves of Engelhardtia roxburghiana Wall. were assessed by determining its scavenging rates against radicals, the reducing capacity, and the effect against lipid membrane oxidation. The optimal extraction process was: ethanol concentration of 64 %, microwave temperature of 56 ℃, and microwave time of 67 s. When the concentration of the alcohol extract from Engelhardtia roxburghiana Wall. was higher than 0.8 mg/mL, its reducing ability and scavenging ability against ABTS+ radicals were obviously stronger than those of astilbin group(P<0.05). In the tested concentration range, the effect of the alcohol extract against lipid membrane oxidation was also significantly higher than that of astilbin(P<0.01).This study shows that response surface methodology is feasible to optimize the alcohol extraction process of the leaves of Engelhardtia roxburghiana Wall. The alcohol extract of the leaves of Engelhardtia roxburghiana Wall. has antioxidant capacity and has potential application in biomedical field.
