摘 要:小麦条锈病一直是影响我国小麦稳产高产的最重要真菌病害之一,防治小麦条锈病最为经济有效的措施就是培育持久抗性品种,鉴别并利用优良抗源是抗病育种的前提。来自美国农业部收集的种质PI 90279和国际玉米小麦改良中心(CIMMYT)的品种Chilero自引进我国后,一直对小麦条锈病保持良好的抗性。前期利用澳大利亚春小麦Avocet S(AvS)作为母本,分别与PI 90279和Chilero杂交构建了F_(2)和重组自交系(RIL)遗传群体,苗期利用CYR34小种接种鉴定后发现,AvS/PI 90279的186个F_(2)群体抗感比为1∶3(χ^(2)=0.58,P=0.27),AvS/Chilero的154个RIL群体抗感比为1∶1(χ^(2)=0.84,P=0.19),说明2个材料的苗期抗性均为一对基因控制。利用集群分离分析(BSA)结合小麦高密度660K芯片策略,对2个群体快速进行基因定位,均在染色体7B相同的位置上成功挖掘出大量的与抗病有关的差异SNP,说明2个抗源的条锈病抗性是由同一个基因控制的。此外,AvS×Chilero群体前期基于DArT标记已构建好全基因组连锁图谱,利用该图谱对苗期抗性表型进行遗传定位,同样也将目标位点锁定在7B染色体上,暂命名为YrCHI。从相对遗传位置、条锈病抗性表现以及育种系谱看,YrCHI可能与YrZM103为同一基因。The improvement of stripe rust resistance in cultivars is viewed to be the most attractive option to control stripe rust.The new emerging race CYR34 along with V26 pathotype group has destroyed most of resistance cultivars.Hence,long-term prevention depends on the availability of genetically diverse sources of resistance.Wheat accessions PI 90279 from the USDA National Small Grains Collection and Chilero from CIMMYT confer adequate resistance to stripe rust both in the United States,Mexico and China for many years.The study dissected the genetic basis of stripe rust resistance on 186 F2 populations produced by crossing Avocet S and PI 90279 and 152 recombinant inbred lines(RILs)from the cross of Avocet S and Chilero at seedling test in the greenhouse.After seedling test,the ratio of resistance to susceptible in F2 population was 1∶3(χ^(2)=0.58,P=0.27)and the ratio of resistance to susceptible in RIL population was 1∶1(χ^(2)=0.84,P=0.15).Bulked segregant analysis coupled with wheat 660K SNP array placed the majority of SNPs differences both on chromosome arm 7BL.These results indicated that resistance in PI 90279 and Chilero might be controlled by the same single dominant gene.In addition,QTL analysis with genome-wide mapping using DArT sequencing in AvS×Chilero RIL population showed that a single gene,temporarily named YrCHI,was also mapped on chromosome 7B.From the relative genetic location,resistance to stripe rust and breeding pedigree,YrCHI may be the same gene as YrZM103.
