低温胁迫下6种马铃薯组培苗耐寒性评价

摘　　要：为研究山西省地区规模化种植且具有代表性的马铃薯品种晋薯16号、冀张薯12号、并薯6号、克新1号、费乌瑞它、大西洋的抗寒性差异,以筛选出耐寒性强的品种,利用低温培养箱对生长28 d左右的马铃薯脱毒组培苗进行4℃和-4℃低温胁迫处理,测定其脯氨酸(Pro)含量、可溶性糖含量、过氧化氢酶(CAT)活性、过氧化物酶(POD)活性4个与耐寒性相关的生理指标;并采用隶属函数法对6个不同品种马铃薯抗寒性进行综合评价和分析。结果表明,随着胁迫温度的降低,6个品种马铃薯的脯氨酸含量整体上呈先上升后下降的趋势,可溶性糖含量呈持续上升的趋势,CAT活性呈持续上升趋势,POD活性呈先上升后下降趋势;晋薯16号、冀张薯12号、大西洋的POD活性的峰值出现在-4℃。采用隶属函数法得出,抗寒性强弱依次为冀张薯12号>并薯6号>晋薯16号>克新1号>大西洋>费乌瑞它。To study the differences in cold resistance of the representative potato varieties Jinshu 16,Jizhangshu 12,Bingshu 6,Kexin 1,Favorita and Atlantic,which were planted on a large scale in Shanxi province variety with strong cold tolerance,to screen out potato varieties with strong cold tolerance.Use a low temperature incubator to treat potato virus-free tissue culture seedlings with a growth cycle of about 28 d under 4℃and-4℃low temperature stress,and determine their proline(Pro)content,soluble sugar content,and catalase(CAT)Activity and peroxidase(POD)activity four physiological indicators related to cold tolerance.The membership function analysis method was used to comprehensively evaluate and analyze the cold resistance of six different potato varieties.The results showed that with the decrease of stress temperature,proline content of the 6 varieties of potato showed an overall upward trend and then a downward trend.Soluble sugar content showed an overall upward trend,CAT activity showed an overall upward trend,POD increased first and then decreased.The peak of POD activity of Jinshu 16,Jizhangshu 12,and Atlantic appeared at-4℃.According to the membership function method,the order of cold resistance manifestation is Jizhangshu 12>Bingshu 6>Jinshu 16>Kexin 1>Atlantic>Favorita.
