尚玺胶囊的抗炎作用及其对血清IL-1β、TNF-α和炎症组织中PGE_(2)的影响

摘　　要：目的评估尚玺胶囊的抗炎作用,探讨其对血清IL-1β、TNF-α和炎症组织中PGE_(2)的影响。方法选择角叉菜胶诱导的大鼠急性炎症模型,测定了大鼠的足周径和足肿胀抑制率。随后,采用生化法测定了血清中超氧化物歧化酶(Superoxide dismutase,SOD)活力、丙二醛(Malondialdehyde,MDA)含量;采用酶联免疫吸附实验(ELISA)检测血清中白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)-含量和炎症足爪中前列腺素E_(2)(PGE_(2))含量。结果相较于对照组,给予尚玺胶囊后,炎症足爪肿胀度在1至4小时内显著降低。当尚玺胶囊的给药剂量为300mg·kg^(-1)时,2小时后足肿胀抑制率达到峰值(7.58%),接近于白藜芦醇的抑制率(8.30%);另外,血清中MDA、IL-1β和TNF-α的含量下降、SOD含量升高,炎症组织中PGE_(2)的含量也显著降低。结论尚玺胶囊在300 mg/kg的剂量下能减轻动物的急性炎症反应,其作用机制可能与降低血清IL-1β、TNF-α水平、减少炎症组织中PGE_(2)合成和抑制组织炎症中脂质过氧化产物的形成有关。Objective To evaluate the anti-inflammatory activity of Shang-Xi Capsule(SXC)and explore its efftct on IL-1β,TNF-αin serum,and PGE_(2)in inflammatory tissues.Methods The carrageenan-induced paw edema experiment was performed to evaluate the anti-inflammatory effect of SXC.And serum superoxide dismutase(SOD),malondialdehyde(MDA),IL-1β,TNF-α and PGE_(2) were measured.Results Compared with the control group,SXC at the dosage of 300 mg·kg^(-1) significantly decreased the swelling of inflammatory paw within 1 to 4 hours after the administration,and the inhibition rate of paw swelling reached a peak(7.58%)after 2 hours,which was close to that of resveratrol(8.30%>).In addition,it can reduce the content of PGE_(2) and NO in inflammatory tissue and the content of NO,MDA,IL-1β,and TNF-α in serum.Conclusion SXC can reduce the acute inflammation induced by carrageenan.Its mechanism of action may be associated with reducing PGE_(2) synthesis in inflammatory tissue,inhibiting the formation of lipid peroxide products in tissue inflammation,and reducing the content of inflammatory factor TNF-α.
