摘 要:目的探究人脐间充质干细胞(hucMSCs)外泌体(hucMSCs exo)对子宫内膜癌细胞生长和转移能力的影响。方法提取并鉴定hucMSCs exo,将ECC1细胞与10μg/mL hucMSCs exo(hucMSCs exo组)以及等体积PBS共孵育,ECC1细胞分别转染si-NC和si-Fascin-1(si-NC组和si-Fascin-1组),ECC1细胞转染Fascin-1质粒的同时与hucMSCs exo孵育(hucMSCs exo+Fascin-1组),MTT法检测ECC1细胞增殖能力,Transwell小室法检测ECC1细胞迁移和侵袭能力,Western blot法检测Fascin-1、LC3-Ⅱ、p62的表达水平。结果差速离心法提取的hucMSCs exo符合外泌体的特征。相比于PBS组,hucMSCs exo组ECC1细胞增殖、迁移和侵袭能力显著降低,Fascin-1表达显著下调,LC3-Ⅱ表达显著上调,p62表达显著下调(P<0.05)。相比于si-NC组,si-Fascin-1组细胞LC3-Ⅱ表达显著上调,p62表达显著下调。hucMSCs exo+Fascin-1组细胞增殖、迁移和侵袭能力显著高于hucMSCs exo组细胞(P<0.05)。结论 hucMSCs exo显著抑制Fascin-1而诱导细胞自噬,进而抑制子宫内膜癌细胞的生长和转移。Objective To explore the effect of human umbilical mesenchymal stem cell(hucMSCs)exosomes(hucMSCs exo) on the growth and metastasis of endometrial cancer cells. Methods Extract and identify the hucMSCs exo. 10μg/mL hucMSCs exo(hucMSCs exo group) and equal volume PBS were co-incubated with ECC1 cells. ECC1 cells were transfected with si-NC and si-Fascin-1(si-NC group and si-Fascin-1 group) respectively. ECC1 cells were transfected with Fascin-1 plasmid and incubated with hucMSCs exo(hucMSCs exo+Fascin-1 group). The proliferation of ECC1 cells was detected by MTT method, and the migration and invasion of ECC1 cells were detected by Transwell chamber method, The expressions of Fascin-1,LC3-Ⅱ and p62 were detected by Western blot. Results The hucMSCs exo extracted by differential centrifugation met the characteristics of exosomes. Compared with the PBS group, the proliferation, migration and invasion ability of ECC1 cells in the hucMSCs exo group was significantly reduced, the expression of Fascin-1 was significantly down-regulated, the expression of LC3-Ⅱ was significantly up-regulated, and the expression of p62 was significantly down-regulated. Compared with the si-NC group, the expression of LC3-Ⅱ in the si-Fascin-1 group was significantly up-regulated, and the expression of p62 was significantly down-regulated. The cell proliferation, migration and invasion ability of hucMSCs exo+Fascin-1 group was significantly higher than that in hucMSCs exo group. Conclusions Human umbilical mesenchymal stem cell exosomes significantly inhibit Fascin-1 and induce autophagy, thereby inhibiting the growth and metastasis of endometrial cancer cells.
