摘  要:目的观察糖尿病大鼠肾小管糖萼标志物多配体蛋白聚糖1(Syndecan, SDC1)表达与肾脏早期损伤的关系。方法 4周龄雌性SD大鼠20只,随机选择6只为对照组,另14只建立糖尿病大鼠模型,将造模成功12只随机分为糖尿病组和干预组,每组6只,干预组尾静脉注射舒洛地特10 mg/(kg·d),持续6周,处死大鼠观察肾脏病理变化,用免疫组织化学、Western blot和实时荧光定量PCR法测SDC1和SDC4表达。结果糖尿病组大鼠肾小球病变轻微,肾小管损伤评分明显高于对照组(P<0.01);干预组肾小管损伤评分低于糖尿病组(P<0.01)。3组SDC4累积吸光度值比较,差异无统计学意义(P>0.05);3组SDC1累积吸光度值比较,差异有统计学意义(P<0.01);糖尿病组大鼠SDC1蛋白及mRNA表达均明显低于对照组(P<0.01);干预组SDC1蛋白及mRNA表达较糖尿病组明显升高(0.44±0.10 vs 0.23±0.08,0.79±0.23 vs 0.43±0.17,P<0.01)。结论糖尿病大鼠肾脏早期病理损伤可能与肾小管SDC1表达减少相关,补充糖胺聚糖可增加肾小管SDC1表达,减轻肾脏病理损伤。Objective To study the relationship between expression of glycocalyx marker SDC1 and early kidney injury in DM rats.Methods Of the 20 female SD rats aged 4 weeks, 6 served as a control group.The 12 rats in the successfully established model were randomly divided into DM group(n=6) and treatment group(n=6).The animals in treatment group were injected with sulodexide through the tail vein(10 mg/kg·d) for 6 weeks and those in DM group were injected with an equal volume of saline.The expressions of SDC1 and SDC4 were detected by immunohistochemical staining, Western blot and qRT-PCR respectively.Results The glomerular injury was mild, the renal tubules injury were severe with massive vacuolar degeneration and brush edge exfoliation.The renal tubule injury score was significantly higher in DM group than that in control group(P<0.01).The glomerular injury and renal tubule injury were significantly milder and the renal tubule injury score was significantly lower in treatment group than that in DM group(P<0.01).The SDC1 was expressed in microvilli of renal tubules and significant difference was detected in accumutive absorbance among the 3 groups(P<0.01).The expression levels of SDC1 protein and mRNA were significantly lower in DM group than that in control group(P<0.01) and significantly higher in treatment group than in DM group(0.44±0.10 vs 0.23±0.08,0.79±0.23 vs 0.43±0.17,P<0.01).Conclusion The early kidney injury is related with the decreased SDC1 expression in DM rats, and sulodexide can upregulate the expression of SDC1 and improve the pathological injury of kidney in DM rats.
