摘 要:目的:研究巫山淫羊藿炮制过程中黄酮类化合物含量的变化规律,并获得符合药典标准的巫山淫羊藿药材最佳炮制工艺。方法:采用高效液相色谱法(HPLC)测定不同温度和受热时间炮制条件下朝藿定A、B、C和淫羊藿苷的含量变化,经方差分析研究不同温度和受热时间对4种黄酮类成分的含量变化影响。结果:不同温度与受热时间对巫山淫羊藿炮制过程中黄酮类成分朝藿定A、B含量变化无显著影响(P>0.05);朝藿定C含量变化受温度影响显著(P<0.05),受热时间对其含量变化无显著影响(P>0.05),在100℃、120℃、140℃、160℃和180℃炮制时含量均呈下降趋势;淫羊藿苷含量受温度和受热时间影响显著(P<0.05),含量整体呈先增后减趋势,在160℃下炮制20min时,淫羊藿苷含量达0.96%,比未炮制时含量提升8.56倍,高于药典标准(0.50%)。结论:为巫山淫羊藿的炮制工艺制定、炮制机理的探索以及临床应用提供科学依据。Objective:To study the variation of flavonoids content during the processing of Epimedium wushanensis,and obtain the best processing technology of Epimedium wushanense that meets the pharmacopoeia standards.Methods:High performance liquid chromatography(HPLC)was used to determine the changes in the content of Epimedium A,B,C and icariin under different temperature and heating time processing conditions.The analysis of variance was used to study the effects of different temperatures and heating times on 4 flavonoids.The content of ingredients changes.Results:Different temperature and heating time had no significant effect on the changes in the content of flavonoids Epimedium A and B during the processing of Epimedium wushanense(P>0.05);the content of Epimedium C was significantly affected by temperature(P<0.05).The content of icariin was not significantly affected by time(P>0.05),and the content showed a downward trend when processed at 100℃,120℃,140℃,160℃and 180℃;the content of icariin was significantly affected by temperature and heating time(P<0.05),the overall content showed a trend of first increasing and then decreasing.When processed at 160℃for 20 minutes,the content of icariin reached 0.96%,which was 8.56 times higher than that without processing,which was higher than the pharmacopoeia standard(0.50%).Conclusion:Provides scientific basis for the formulation of processing technology,the exploration of processing mechanism and clinical application of Epimedium wushanense.
