摘  要:为明确含DEV-gB基因重组减毒沙门氏菌在鸭免疫器官中的分布动态,本研究将重组减毒沙门氏菌口服接种鸭(即重组细菌组),同时设立重组质粒组、弱毒疫苗组和空白对照组,于免疫后第7、14、21、28、35、42和49天时,捕杀试验鸭,采集鸭脾脏、胸腺、法氏囊和哈德氏腺等免疫器官,采用间接免疫荧光法进行DEV-gB抗原检测观察,结果显示:空白对照组鸭免疫器官在7个时间点均未检测到DEV-gB抗原信号;免疫后,重组细菌组、弱毒疫苗组和重组质粒组鸭免疫器官均能检测到DEV抗原阳性信号,但含DEV抗原阳性信号细胞数量从多到少的组别顺序是弱毒疫苗组、重组细菌组和重组质粒组,且重组细菌组与弱毒疫苗组差异不显著(P>0.05);检出含DEV抗原阳性信号的免疫器官先后顺序是脾脏、法氏囊、哈德氏腺和胸腺,而含DEV抗原阳性信号细胞数量从多到少的免疫器官顺序是脾脏、法氏囊、胸腺和哈德氏腺。由此可见,本研究构建的携带DEV-gB基因重组减毒沙门氏菌能在鸭免疫器官中得到良好的表达,为该重组细菌疫苗的临床应用奠定了理论基础。To determine the distribution dynamics of recombinant attenuated salmonella carring DEV-gB gen in the immune organs of ducks,and inoculated with recombinant bacteria,recombinant plamid,attenuated vaccine and sterile PBS silution(blank control),respectively.The experimental ducks were killed at the 7^(th),14^(th),21^(th),28^(th),35^(th),42^(th) and 49^(th) day after immunization,and the spleen,thymus,Fabricius bursa and Hardy's gland were collected,and detected for DEV-gB antigen by indirect immunofluorescence method.The results showed that no signal of DEV-gB antigen was detected in duck immune organs of blank control group at every time-point,but positive signal presented in the other three groups.And the cell number with DEV-gB positive signal in the recombinant bacteria group was,the most,followed by attenuated vaccine group and recombinant plasmid group,but there was no significant difference between the recombinant bacteria group and the attenuated vaccine group.The order of the immune organs detected with positive signal of DEV-gB antigen were respectively spleen,Fabricius bursa,Hardy's gland and thymus,while the cell number with DEV-gB positive signal in spleen were the most,followed by Fabricius bursa,thymus and Hardy's gland.Therefore,the recombinant attenuated Salmonella carrying DEV-gB gene could be well expressed in duck immune organs,laying a theoretical basis for the clinical application of the recombinant bacterial vaccine.
