摘 要:目的研究别隐品碱对小鼠心房肌细胞的T-型钙电流的影响。方法采用胶原酶Ⅱ急性消化分离小鼠心房细胞,别隐品碱被用灌流法作用于细胞,应用电压钳模式测量小鼠心房肌细胞T-型钙电流(I_(Ca,T))。结果在细胞外应用30μM别隐品碱能降低小鼠心房肌细胞I_(Ca,T)电流密度。在-40 mV电压下,使其峰值密度从(3.3±0.4)pA/pF减少至(1.7±0.2)pA/pF(n=15,P<0.01)。门控机制检测结果显示,别隐品碱可使I_(Ca,T)通道电流的稳态激活过程发生改变,曲线正向移动,使其失活后恢复时间常数(τ)延长,从(54.9±3.3)ms变为(75.6±3.4)ms(n=15,P<0.01)。揭示别隐品碱的作用机制可能通过抑制I_(Ca,T)稳态激活和失活后恢复过程所致。结论别隐品碱可降低小鼠心房肌细胞T-型钙电流密度,可能与其延迟通道激活及失活后再激活有关。OBJECTIVE To study the effect of allocryptine on T-type calcium currents in mouse atrial myocytes.METHODS Collagenase and protease were used to digest and isolate mouse atrial myocytes.Allocryptine was applied to the cells by perfusion,and the whole-cell patch clamp method was used to record the T-type calcium current(I_(Ca,T))of mouse atrial myocytes.RESULTS The extracellular application of 30μM allocryptine can reduce the density of I_(Ca,T)current of mouse atrial myocytes.Under-40 mV voltage,its peak density was reduced from(3.3±0.4)pA/pF to(1.7±0.2)pA/pF(n=15,P<0.01).The study on the gating mechanism of T-type calcium channels found that allocryptine could shift the steady-state activation curve of I_(Ca,T)to the right,and extended the recovery time constant after inactivation(τ)from(54.9±3.3)ms to(75.6±3.4)ms(n=15,P<0.01).The results of the study revealed that allocryptine mainly reduced the density of I_(Ca,T)by inhibiting the steady-state activation and recovery of the channel after inactivation.CONCLUSION Allocryptine can reduce the T-type calcium current of mouse atrial myocytes,which may be related to the delayed channel activation and reactivation after inactivation.
