摘 要:目的分析探讨酪氨酸蛋白激酶受体(EphB4)、自噬基因(Beclin-1)在稽留流产绒毛组织中的表达及与微血管密度的相关性。方法收集2020年1月至2020年12月在南通大学附属医院妇产科确诊为稽留流产终止妊娠的患者62例为观察组,另选取同期正常早孕自愿终止妊娠患者60例为对照组;采用蛋白质印迹法检测两组绒毛组织中EphB4、Beclin-1蛋白的表达,采用免疫组化法检测两组绒毛组织中微血管密度(MVD),并分析EphB4、Beclin-1表达水平与MVD的相关性。结果观察组绒毛组织中EphB4蛋白相对表达量明显低于对照组(t=4.97,P<0.01);观察组绒毛组织中Beclin-1蛋白相对表达量高于对照组,差异有统计学意义(t=23.04,P<0.01);观察组绒毛组织中MVD明显低于对照组(t=12.42,P<0.01);在稽留流产患者绒毛组织中,EphB4蛋白相对表达量与MVD呈正相关(r=0.44,P<0.01),Beclin-1蛋白相对表达量与MVD呈负正相关(r=-0.52,P<0.01)。结论绒毛组织中EphB4、Beclin-1水平与稽留流产患者MVD有相关性,早孕期EphB4的低表达和Beclin-1水平高表达可导致血管生成障碍。Objective To analyze expression of tyrosine protein kinase receptor erythropoietin producing hepatoma cell line subset EphB4 and autophagy gene(Beclin-1) in missed abortion chorionic villous tissues and their correlations with microvessel density(MVD).Methods From January 2020 to December 2020,62 patients who were diagnozed as missed abortion and underwent induced abortion in Department of Gynecology and Obstetrics, The Afilliated Hospital of Nantong University were selected as study subjects and divided into observation group, and other 60 normal early pregnant women who accept induced abortion voluntarily in the same period were selected as the control group.Western blotting was used to detect expressions of EphB4 and Beclin-1 proteins in the chorionic villous tissues of the pregant women in the two groups, and immunohistochemical method was used to detect MVD in the chorionic villous tissues of the pregnant women in the two groups.And correlations of the expression levels of EphB4 and Beclin-1 with MVD were analyzed.Results The relative expression of EphB4 protein in the chorionic villous tissues of the pregnant women in the observation group was lower than that of the pregnant women in the control group(t=4.97,P<0.01),while the relative expression of Beclin-1 protein in the chorionic villous tissues of the pregnant women in the observation group was significantly higher than that of the pregnant women in the control group(t=23.04,P< 0.01).MVD in the chorionic villous tissues of the pregnant women in the observation group was significantly lower than that of the control group(t=12.42,P<0.01).In the chorionic villous tissues of those pregnant women in the observation group, the relative expression of EphB4 protein was positively correlated with MVD(r=0.44,P<0.01),while the relative expression of Beclin-1 protein was negatively correlated with MVD(r=-0.52,P<0.01).Conclusion The expression levels of EphB4 and Beclin-1 in the chorionic villous tissues are correlated with the MVD in the chorionic villous tissues of
