摘 要:对采集自湖北省十堰市房县地区的柠檬鳞伞(Pholiota limonella)子实体进行组织分离,并对获得的菌株进行分子鉴定、生物学特性及驯化栽培研究。结果表明:在实验范围内,柠檬鳞伞菌丝生长的最适温度为25℃,最适pH为7.0,最适碳源为麦芽糖和蔗糖,最适氮源为硝酸钾。使用木屑为主要培养料栽培柠檬鳞伞,菌袋接种后于(25±1)℃、黑暗条件下培养,约60 d后菌丝满袋;出菇时的环境温度为10~22℃,相对湿度>90%,光照0~125 lx,开袋后25~30 d形成原基,35~40 d即可采收;平均生物学效率为24.01%。A wild Pholiota limonella fruiting body collected from Fangxian area of Hubei Province was subjected to tissue separation and then identified by molecular markers,studied for biological characteristics and domestication.The results showed that the optimal temperature,pH,carbon source and nitrogen source for mycelial growth were 25℃and 7.0,maltose/sucrose and potassium nitrate,respectively.Using saw dust as the major cultivation substrate material,mycelia fully filled a cultivation bag after cultivation in darkness at(25±1)℃for 60 d.During fruiting body development,the appropriate environmental temperature and illumination condition were 10-22℃and 0-125 lx,respectively.About 25-30 d after bag opening,primordia started to appear and fruiting bodies were harvested around 35-40 d.The average biological efficiency was 24.01%.
