摘 要:采用病证结合方法,构建三种证型肺气虚、脾气虚、肺脾气虚哮喘小鼠模型,研究三种证型对哮喘小鼠特异性/非特异性免疫功能及气道炎症的影响及关联性.与空白对照组相比,脾气虚、肺脾气虚哮喘小鼠T·B淋巴细胞增殖功能降低、T细胞亚型CD3+、CD4+占比减少、IgG、IgA、IgM表达水平下降(P<0.05);且三种证型哮喘小鼠BALF中各类炎性细胞计数、分泌的细胞因子IL-4、IL-5、IL-13含量增多,且肺脾气虚哮喘组气道阻力上升至2倍(P<0.05).模型组之间相比,肺脾气虚哮喘组较单纯哮喘组和肺气虚哮喘组T淋巴细胞增殖功能降低,T细胞亚型CD3+、CD4+比例下降,体液免疫指标IgA、IgM含量下降明显,且肺脾气虚哮喘小鼠BALF中嗜酸性粒细胞增加、血清中OVA-IgE浓度高于单因素证候模型组(P<0.05).结果表明:三种证型对哮喘小鼠特异性/非特异性免疫功能及气道炎症有影响且具有一定关联性.The combination of disease and syndrome was used to construct three syndromes of asthma mouse models to study the effects of the three syndromes including spleen-qi deficiency asthma,lung-qi deficiency asthma and lung-spleen qi deficiency asthma.Compared with the blank control group,immune organ index,T·B lymphocyte proliferation,T cell subtype CD3+,CD4+proportion,humoral immune index IgG,IgA,IgM content in the asthma group with deficiency of spleen-qi and asthma group with deficiency of lung spleen qi decreased(P<0.05).IL-4,IL-5,and IL-13 in the BALF of the three types of asthma groups increased,and the airway resistance in the asthma group with the deficiency of lung-spleen-qi increased to 2 times of the blank group(P<0.05).Compared with the model groups,the deficiency of lung-spleen qi asthma group was lower than the simple asthma group and the lung-qi deficiency asthma group in terms of immune organ index,T lymphocyte proliferation,T cell subtype CD3+,CD4+ratio,humoral immune index content,and deficiency of lung-spleen qi asthma group.The concentrations of eosinophils and serum OVA-IgE in BALF were higher than those in the single-factor syndrome model group(P<0.05).The results show that the three types of syndromes have effects on specific/non-specific immune function and airway inflammation in asthmatic mice and have certain correlations.
