摘 要:目的探讨菟丝子、女贞子治疗绝经后骨质疏松症(PMOP)的作用机制。方法检索GeneCards、人类孟德尔遗传数据库(OMIM)数据库确定PMOP相关靶点,然后通过中药系统药理学数据库与分析平台(TCMSP)数据库筛选菟丝子、女贞子中活性化学成分和对应靶点,并通过R语言筛选出药物与疾病的共同靶点,构建“药物-候选化合物-靶点-疾病”网络图。使用蛋白质相互作用网络功能富集分析(String)数据库和Cytoscape软件构建蛋白质互作网络(PPI),应用Autodock vina进行分析对接,使用R语言对菟丝子、女贞子治疗PMOP的关键靶点进行基因本体(GO)和京都基因和基因组百科全书(KEGG)富集分析,确定菟丝子、女贞子治疗PMOP的核心靶点和作用机制。结果共获得菟丝子活性化合物11个,女贞子活性化合物13个,菟丝子、女贞子-PMOP交集靶点113个。与疾病靶标相关的活性成分有15个,菟丝子、女贞子主要参与调控白细胞介素(IL)-6、RAC-α丝氨酸/苏氨酸蛋白激酶、血管内皮生长因子A、表皮生长因子等关键靶标及糖尿病并发症的晚期糖基化终末产物-晚期糖基化终末产物受体信号通路、流体剪切应力和动脉粥样硬化信号通路、IL-17、肿瘤坏死因子、缺氧诱导因子1、内分泌抵抗、细胞凋亡信号通路发挥治疗PMOP的作用。结论菟丝子、女贞子治疗PMOP的作用机制呈多靶点、多系统的特性,除影响骨代谢相关途径外还可影响体内多种代谢途径。Objective To explore the key targets and action mechanisms of Cuscuta chinensis and Ligustrum lucidum in preventing and treating postmenopausal osteoporosis(PMOP).Methods By searching the databases of GeneCards,Online Mendelian Inheritance in Man(OMIM),the targets related to PMOP were selected,and the active chemical components and corresponding targets in Cuscuta chinensis and Ligustrum lucidum were screened through the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and the drugs were screened for the common target of the disease through R language to construct a network diagram of"drug-candidate compound-target-disease".Protein-protein interaction networks functional enrichment analysis(String)database and Cytoscape software were applied to construct protein-protein interaction network(PPI),analysis and docking was done with Autodock vina,and R language was used to perform gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis on the key targets of Cuscuta chinensis and Ligustrum lucidum in the treatment of PMOP,and investigate the core targets and mechanism of action of Cuscuta chinensis and Ligustrum lucidum in the treatment of PMOP.Results A total of 11 active compounds from Cuscuta chinensis,13 active compounds from Ligustrum lucidum,and 113 disease targets from the intersection of Cuscuta chinensis,Ligustrum lucidum and PMOP were obtained.There are 15 active components related to the disease targets,which are mainly involved in the regulation of interleukin(IL)-6,RAC-αserine/threonine-protein kinase,vascular endothelial growth factor A,epidermal growth factor and other key targets,as well as the advanced glycation end products-receptor for advanced glycation end products signaling pathway in diabetic complications,fluid shear stress and atherosclerosis signal pathway,IL-17,tumor necrosis factor,hypoxia-inducible factor 1,endocrine resistance,and apoptosis signal pathways to treat PMOP.Conclusion The mechanism of action of Cuscuta chin
