摘 要:为寻求灵敏有效的柑橘病原物检测技术,建立柑橘黄龙病菌(CLas)和黄化脉明病毒(CYVCV)同时检测的技术。采用双重PCR检测体系,根据CLas的16S和CYVCV的外壳蛋白(CP)基因序列,分别设计2对特异性引物。通过双重PCR技术,能有效检测柑橘是否含有CLas和CYVCV,同时排除假阳性和假阴性。灵敏度分析结果显示,在样品cDNA原液稀释到10^(4)倍时,仍可以检测到2种病原物扩增出的目的条带。此方法能够快速准确地检测柑橘CLas和CYVCV,且具有更高的灵敏度,可为柑橘病原物检测提供便利。To explore the rapid,sensitive and effective detection techniques of citrus pathogens,the identification techniques of citrus Candidatus Liberibacter asiaticus(CLas)and Citrus yellow vein clearing virus(CYVCV)were established.Two pairs of specific primers were designed according to the CLas 16S and the CYVCV coat protein(CP)gene sequence by a dual detection system.By PCR dual technology,can effectively detect whether citrus contains CLas and CYVCV,while excluding false positive and negative.Sensitivity analysis showed that the target sample cDNA solution was diluted to 10^(4) times,the target band amplified by 2 pathogens could still be detected.This method can quickly and accurately detect citrus CLas and CYVCV,and has a higher sensitivity to facilitate the of citrus pathogens.
