摘 要:目的建立一种基于荧光微球的定量免疫层析检测试纸卡,用于农药醚菊酯(etofenprox)的快速定量检测,并对所制备的试纸卡性能进行评价。方法采取竞争抑制免疫层析模式,通过优化试纸各要素,研制醚菊酯检测试纸,并对其标准曲线、检测限、定量限、特异性、准确度、精密度等方面进行性能评价。结果所制备的试纸卡标准曲线回归方程R;值达0.980 1,检测限达1.057 ng/ml;定量限达1.890 ng/ml;与类似农药交叉反应率小于0.10%;添加回收率为98.60%~106.50%;批内和批间变异系数均小于10.00%。结论所研制的醚菊酯荧光定量免疫层析试纸卡各项指标均达到要求,具有特异性好、灵敏度高、检测快速、简便的优点,可以应用于醚菊酯残留检测。Objective To establish a quantitative immunochromatographic test card based on fluorescent microspheres for the rapid and quantitative detection of the pesticide etofenprox, and to evaluate the performance of the test strip card.Methods Based on the competitive immunosuppressive immunochromatographic assay model, and by optimizing the test strip card elements, etofenprox test paper was developed.The performance of the card was evaluated in terms of its standard curve, detection limit, quantitative limit, specificity, accuracy, precision, etc.Results The regression equation R;value of the standard curve of the test card reached 0.980 1,the detection limit reached 1.057 ng/ml, the quantitative limit reached 1.890 ng/ml.The cross-reaction rate with similar pesticides was less than 0.10%,the recovery rate was 98.60%-106.50%,and the coefficient of variation was less than 10.00% within and between batches.Conclusion The indexes of the developed etofenprox fluorescent quantitative immunochromatographic test strip card meet the requirements, and has the advantages of good specificity, high sensitivity, fast and simple detection, and can be applied to the detection of etofenprox residues.
