摘 要:目的观察NLRC5在大鼠腰椎椎板切除术后硬膜外瘢痕形成中的表达情况。方法将60只3月龄雄性SD大鼠随机分为手术组和假手术组,每组30只,手术组大鼠行腰椎椎板切除术,假手术组暴露椎板后不做特殊处理。模型建立4周后处死动物获取硬膜外组织标本,采用Masson染色评价组织病理学改变情况,采用实时荧光定量PCR检测NLRC5、结缔组织生长因子(CTGF)、α-平滑肌肌动蛋白(α-SMA)和Ⅰ型胶原(COLⅠ)的mRNA表达水平,蛋白质印迹法检测NLRC5、CTGF、α-SMA蛋白表达水平,ELISA法检测COLⅠ蛋白含量。结果Masson染色结果显示,手术组大鼠硬膜外组织结构破坏,可见大量纤维细胞,假手术组大鼠硬膜外组织结构完整。实时荧光定量PCR检测结果显示,手术组大鼠硬膜外组织NLRC5、CTGF、α-SMA和COLⅠ的mRNA表达高于假手术组,差异有统计学意义(P<0.05)。蛋白质印迹检测结果显示,手术组大鼠硬膜外组织NLRC5、CTGF和α-SMA的蛋白表达高于假手术组,差异有统计学意义(P<0.05)。ELISA检测结果显示,手术组大鼠硬膜外组织COLⅠ的含量高于假手术组,差异有统计学意义(P<0.05)。结论NLRC5可能参与大鼠腰椎椎板切除术后硬膜外瘢痕形成的发展过程。Objective To investigate the role of NLRC5 in epidural scar formation after laminectomy in rats.Methods Sixty 3-month-old male SD rats were randomly divided into the operation group and sham operation group(30 in each group).Lumbar laminectomy was performed in the operation group and no special treatment was done after exposure of the lamina in the sham-operation group.Four weeks after the establishment of the model,the animals were killed to obtain epidural tissue samples.Histopathological changes of epidural tissue in the 2 groups were evaluated by Masson staining.The mRNA of NLRC5,connective tissue growth factor(CTGF),α-smooth muscle actin(α-SMA)and collagenⅠ(COLⅠ)were analyzed by real-time quantitative PCR,and protein expressions of NLRC5,CTGF andα-SMA were detected by Western blotting.The content of COLⅠwas detected by ELISA.Results Masson staining showed that the epidural tissue structure was destroyed and a lot of fibrous cells were seen in the operation group,while the epidural tissue structure was intact in the sham operation group.The expressions of NLRC5,CTGF,α-SMA and COLⅠmRNA of the epidural tissue in the operation group were significantly higher than those in the sham operation group,with a statistical significance(P<0.05).The expressions of NLRC5,CTGF andα-SMA protein of the epidural tissue in the operation group were significantly higher than those in the sham operation group,with a statistical significance(P<0.05).The COLⅠcontent of the epidural tissue in the operation group was higher than that in the sham operation group,with a statistical significance(P<0.05).Conclusion NLRC5 may be involved in the progression process of epidural scar formation after lumbar laminectomy.
