摘 要:迄今为止,新冠病毒肺炎疫情已造成全球近2亿人感染,数百万人因新冠肺炎而罹难。针对疫情病原——新型冠状病毒,人们对其重要药物靶标开展了深入而详实的研究。对药物靶标蛋白三维结构的解析,对于我们了解药物作用机制,以及开发新的药物具有非常重要的指导意义。文章总结了针对新冠病毒主蛋白酶及转录复制复合体的多种结构,分析了药物小分子抑制相应靶标蛋白的机制,以及对于抗击新冠疫情的重要意义。So far,more than 200 million people were infected and millions of people have died during this worldwide COVID-19 pandemic.In response to the SARS-CoV-2,the pathogen of the epidemic,scientists have carried out in-depth and detailed researches on its drug targets.The analysis of the three-dimensional structures of drug target proteins has guiding significance for people to understand the mechanism of drug candidates and develop new drugs.This article summarizes the structures of the main protease and transcription-replication complex of SARS-CoV-2,analyzes the mechanism of small molecules inhibiting the corresponding target protein,and elucidates the significance in fighting the COVID-19 pandemic.
