摘  要:目的探讨木犀草素对急性尿路感染(UTI)大鼠的改善作用及相关机制。方法将40只大鼠随机分为对照组、模型组、木犀草素低剂量组(10mg/kg)、木犀草素中剂量组(20 mg/kg)和木犀草素高剂量组(40 mg/kg),除对照组外,其他组大鼠建立急性UTI大鼠模型,模型.建立后对各组给药大鼠进行灌胃给药,对照组和模型组大鼠仅给予等量生理盐水,连续2周,期间观察各组大鼠的尿量、尿白细胞数量、膀胱指数;采用HE染色观察各组大鼠膀胱组织的病理损伤情况;ELISA试剂盒检测各组大鼠的血清白介素6(IL6)、白介素-8(IL-8)含量;Western blot检测各组大鼠膀胱组织中Toll样受体4(TLR4)、p65、p-p65蛋白表达情况。结果与对照组相比,模型组大鼠的尿量减少,尿白细胞数目显著增多,膀胱指数增加(P<0.05);与模型组相比,木犀草素中、高剂量组大鼠的尿量增加(P<0.05),尿白细胞数目显著减少(P<0.05),膀胱指数下降(P<0.05)。对照组大鼠膀胱组织无炎性浸润,组织结构正常无病变。模型组大鼠膀胱组织充血水肿存在大量中性粒细胞炎性浸润、部分黏膜上皮细胞脱落,与模型组相比,木犀草素低剂量组大鼠的病理切片变化无显著区别,木犀草素中、高剂量组大鼠膀胱组织病理情况得到不同程度的改善。与对照组比较,模型组大鼠膀胱组织的IL-6、IL-8含量增加,TLR4、p-p65蛋白表达升高(P<0.05);与模型组比较,木犀草素中、高剂量组大鼠膀胱组织IL-6、IL-8含量下降,TLR4、p-p65蛋白表达降低(P<0.05)。结论木犀草素能够抑制急性UTI大鼠的炎症反应,使其症状得到改善,其机制可能与木犀草素抑制TLR4/NF-κB信号通路有关。Objective To investigate the improving effect of luteolin on rats with acute urinary tract infection(UTI)and related mechanisms.Methods Forty rats were randomly divided into control group,model group,luteolin low-dose group(10 mg/kg),luteolin medium-dose group(20 mg/kg)and luteolin high-dose group(40 mg/kg).Except for the control group,rats in other groups were established with acute UTI rat models.After the model was established,rats in each administration group were given intragastric administration.The rats in the control group and the model group were given the same amount of physiological salt water for 2 consecutive weeks,during which the urine output,the number of white blood cells in urine and the bladder index of each group were observed.The pathological damage of the bladder tissue of each group was observed by HE.The serum IL-6 and IL-8 content of each group were detected by ELISA kit.Western-blot detected the expression of TLR4,p65,p-p65 protein in the bladder tissue of each group of rats.Results In the control group,there was no inflammatory infiltration in the bladder tissue,normal tissue structure and no lesions.In the model group,the bladder tissue was congested and edematous,there was a large number of inflammatory infiltration of neutrophils,some mucosal epithelial cells were fell off,the urine volume decreased,and urine number of white blood cells increased significantly,and the bladder index increased(P<0.05).The pathological conditions of the bladder tissue of rats in the middle and high doses of luteolin were improved to varying degrees,and the urine output increased,the number of white blood cells in urine decreased significantly,and the bladder index decreased(P<0.05).Compared with the control group,the IL-6 and IL-8 levels in the bladder tissue of the model group increased,and the expression of TLR4,p65,p-p65 protein increased(all P<0.05).Compared with the model group,the contents of IL-6 and IL-8 and the expression of TLR4,p65,and p-p65 proteins were decreased in the bladder tissue of lu
