摘 要:目的研究高碘饮水后ClC-3对小鼠甲状腺功能调节的作用。方法SPF级FVB小鼠野生型(3月龄,16只)和ClC-3转基因小鼠(3月龄,16只)各分为对照组(Cont组)和高碘组(NaI组)。HE染色比较小鼠甲状腺组织学形态改变;免疫组化和Western blotting观察小鼠甲状腺ClC-3的表达与分布;ELISA检测小鼠血清游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)和促甲状腺激素(TSH)的浓度。结果与WT小鼠相比,ClC-3转基因小鼠甲状腺滤泡上皮细胞增生,滤泡腔数目增加且体积扩大,FT3、FT4分泌增加;高碘饮水后,WT小鼠和ClC-3转基因小鼠ClC-3蛋白表达增加;甲状腺细胞增生,伴滤泡腔数目增多和体积扩大;FT3和FT4明显增高(P<0.05);WT小鼠伴有TSH下降,转基因小鼠无明显改变。结论高碘饮水可使甲状腺ClC-3蛋白表达增加,ClC-3蛋白过表达可导致甲状腺增生和甲状腺激素分泌增加。提示ClC-3参与了碘离子对甲状腺功能的调节过程。Objective To investigate the role of ClC-3 in thyroid function regulated by iodine in mice.Methods Wild type FVB mice(3 month old,n=16)and ClC-3 transgene mice(3 month old,n=16)were selected into control group and iodine-treated group,respectively.HE staining was used to analyze the histological difference of thyroid tissue;Western blotting and Immunohistochemistry were applied to detect the expression and distribution of ClC-3;ELISA assay was used to detect the serum concentrations of free triiodothyronine(FT3),free thyroxine(FT4)and thyrotropin(TSH).Results Compared to WT mice,there were significant proliferation of thyroid follicular cells,accompanied with increased number and size of thyroid antral and higher level of FT3 and FT4,in ClC-3 transgene mice.After drinking high iodine water,the expression of ClC-3 and levels of FT3 and FT4 in both WT mice and ClC-3 transgenic mice increased obviously(P<0.05).Moreover,the level of TSH decreased in WT mice;but there was no significant difference in ClC-3 transgenic mice.Conclusion Over-iodine drinking could increase the expression of ClC-3,induce the proliferation of thyroid follicular cells and increase thyroid hormone level,indicating that ClC-3 could be involved in thyroid function regulated by iodine.
