摘  要:目的研究肺炎支原体感染新生大鼠补体激活水平及免疫因子表达水平。方法随机取新生SD大鼠42只,制备新生大鼠急性肺损伤模型,分为空白组、对照组、模型组(M1、M2、M3、M4、M5组),观察不同感染时长模型组大鼠健康状况,测定外周血各类炎症细胞数目,观察病理状况,酶联免疫法测定肺组织相关免疫因子表达水平、外周血补体蛋白(C3a、C4a、C5a)表达水平。结果模型组大鼠精神状态下降,体质量下降(P<0.05),肺组织病理损伤明显、炎性细胞浸润;模型组大鼠外周血炎症细胞计数高于空白组,支原体包涵体滴度随着感染时间增加而增加(P<0.05);模型组肺组织中白细胞介素(IL)-6、IL-12、IL-17A、转化生长因子β1(TGF-β1)、肿瘤坏死因子α(TNF-α)蛋白表达水平较空白组、对照组升高(P<0.05);模型组外周血中C3a、C4a、C5a补体激活水平远超出空白组、对照组补体激活水平(P<0.05)。结论肺炎支原体感染新生大鼠后肺组织病理变化与肺组织中IL-6、IL-12、IL-17A、TGF-β1、TNF-α蛋白表达水平、补体激活水平存在相关性,随着感染时间增加,肺组织被破坏程度受补体激活水平影响。OBJECTIVE To study the levels of complement activation and immune factors in neonatal rats infected with Mycoplasma pneumoniae. METHODS Totally 42 neonatal SD rats were randomly selected so as to prepare acute lung injury models and were divided into the blank group and the model groups( group M1, M2, M3, M4, M5). The health status of the model groups was observed, the counts of inflammatory cells in peripheral blood were determined, the pathological status was observed, and the expression levels of lung tissue-related immune factors and peripheral blood complement proteins( C3 a, C4 a, C5 a) were detected by enzyme-linked immunosorbent assay. RESULTS The mental state of the rats in the model groups declined, and the body weights were decreased(P<0.05), the rats had obvious pathological damage of lung tissues and infiltration of inflammatory cells. The counts of inflammatory cells in peripheral blood were significantly greater in the model groups than in the blank group, and the titers of Mycoplasma inclusion bodies were significantly increased with time(P<0.05). The expression levels of interleukin(IL)-6, IL-12, IL-17 A, transforming growth factor β1(TGF-β1) and tumor necrosis factor α(TNF-α) in lung tissues were significantly higher in the model groups than in the blank group and the control group(P<0.05). The activation levels of peripheral blood C3 a, C4 a and C5 a complements of the model groups were significantly higher than those of the blank group and the control group(P<0.05). CONCLUSION There is association between the pathological changes of lung tissues of the neonatal rats infected with M.pneumoniae and the expression levels of IL-6, IL-12, IL-17 A, TGF-β1 and TNF-α in lung tissues as well as the activation levels of complements. With the extension of time, the degree of damage of lung tissue is affected by the activation level of complements.
