摘  要:目的研究左氧氟沙星联合蒲公英提取物对细菌性乳腺炎小鼠免疫功能的调节作用。方法采用哺乳期在10~15 d的Balb/c小鼠50只,分为空白对照组、模型对照组、左氧氟沙星组、蒲公英提取物组、左氧氟沙星+蒲公英提取物组,对五组小鼠乳腺组织含菌量、T细胞体外增殖、免疫分型以及细胞周期分布进行分析,同时测定肿瘤坏死因子-α(TNF-α)、皮质醇、减数二次分裂(MII)、外周血中性粒细胞(PMN)吞噬率和胸腺指数。结果相比左氧氟沙星组和蒲公英提取物单独用药组,左氧氟沙星+蒲公英提取物组小鼠乳腺组织中的细菌含量降低(P<0.05),T细胞体外增殖水平降低(P<0.05),T细胞CD25、CD69、CD71表达降低(P<0.05),同时,TNF-α、皮质醇、MII、PMN吞噬率和胸腺指数增加,差异具有统计学意义(P<0.05)。结论左氧氟沙星与蒲公英提取物联合给药显著提高抗菌效果,同时通过调节T细胞增殖和活化水平调节机体的免疫功能,发挥治疗细菌性乳腺炎的功能。OBJECTIVE To observe the effect of levofloxacin combined with dandelion extract on regulating the immune function of the mice with bacterial mastitis. METHODS A total of 50 BALB/c mice with the suckling period in 10-15 days were divided into the blank control group, the model control group, the levofloxacin group, the dandelion extract group, and the levofloxacin plus dandelion extract group. The bacterial colony counts, in vitro proliferation of T cells, immunotypes and cellular cycles were observed and compared among the five groups of mice, meanwhile, the tumor necrosis factor-α(TNF-α), cortisol, meiotic secondary division(MII), phagocytosis rate of peripheral blood neutrophils(PMN) and thymus index were determined. RESULTS As compared with the levofloxacin group and the dandelion extract group, the bacterial colony counts in mammary tissues of the levofloxacin plus dandelion extract group were significantly reduced(P<0.05), the level of in vitro proliferation of T cells was reduced significantly(P<0.05), the expression levels of T cell CD25, CD69 and CD71 were significantly reduced(P<0.05), meanwhile, the TNF-α, cortisol, MII, phagocytosis rate of PMN and thymus index were increased(P<0.05). CONCLUSION Levofloxacin combined with dandelion extract can significantly improve the antibacterial effect and regulate the immune function by regulating the proliferation and activation level of the T cells, showing the effect on treatment of bacterial mastitis.
