摘  要:目的:研究针刺对超负荷运动致大鼠骨骼肌损伤氧化应激损伤、骨骼肌细胞凋亡的影响并探讨相关机制。方法:将60只大鼠随机分成健康对照组、针刺组、超负荷组和超负荷+针刺组,每组15只。检测各组大鼠血清中肌酸激酶(CK)、乳酸脱氢酶(LDH)以及骨骼肌中丙二醛(MDA)、超氧化物歧化酶(SOD)、琥珀酸脱氢酶(SDH)和谷胱甘肽过氧化物酶(GSH-Px)的水平;HE染色观察骨骼肌损伤情况;流式细胞术检测各组大鼠骨骼肌细胞的凋亡率;通过免疫印迹法检测各组大鼠骨骼肌PI3K/AKT信号通路相关蛋白表达情况。结果:与健康对照组比较,超负荷组CK、LDH和MDA水平、骨骼肌细胞凋亡率、Bax蛋白均显著升高(均P<0.05),SOD、SDH、GSH-Px和Bcl-2均显著降低(均P<0.05)。超负荷+针刺组各病理程度明显减轻。与健康对照组比较,超负荷组PI3K和AKT蛋白水平均显著降低(均P<0.05);与模型组比较,超负荷+针刺组PI3K和AKT蛋白水平均显著升高(均P<0.05)。结论:针刺可能通过激活PI3K/AKT信号通路,抑制氧化应激反应和骨骼肌细胞的凋亡,从而缓解大鼠骨骼肌的损伤。Objective:To investigate the effects of acupuncture on oxidative stress injury and apoptosis of skeletal muscle cells in rats induced by exercise overload and explore the related mechanisms.Methods:A total of 60 rats were randomly divided into healthy control group,acupuncture group,overload group and overload+acupuncture group.The levels of creatine kinase(CK) and lactate dehydrogenase(LDH) in serum,and the contents of malondialdehyde(MDA),superoxide dismutase(SOD),succinate dehydrogenase(SDH) and glutathione peroxidase(GSH-PX) in skeletal muscles were detected.Skeletal muscle injury was observed by HE staining.The apoptosis rate of skeletal muscle cells was detected by flow cytometry,and the expression of PI3 K/AKT signaling pathway related proteins in skeletal muscles was detected by western blotting.Results:Compared with healthy control group,the levels of CK,LDH and MDA,the apoptosis rate of skeletal muscle cells and Bax protein level in overload group were significantly increased,while the levels of SOD,SDH,GSH-Px and Bcl-2 were significantly decreased(P<0.05).The pathological degree of overload + acupuncture group was significantly reduced(P<0.05).Compared with healthy control group,the protein levels of PI3 K and AKT in overload group were significantly decreased(P<0.05).Compared with model group,the protein levels of PI3 K and AKT in overload+acupuncture group were significantly increased(P<0.05).Conclusion:Acupuncture may inhibit oxidative stress response and apoptosis of skeletal muscle cells by activating PI3 K/AKT signaling pathway,thus alleviating skeletal muscle injury in rats.
