摘 要:本文分析总结了40年来水仙属植物组织培养与植株再生的研究结果.总结出共性的技术要求:(1)水仙最有效率的离体快繁外植体是“带鳞茎盘的双鳞片”或者是“双鳞片”;(2)采用带鳞茎盘的鳞片或单纯的鳞片诱导愈伤组织,以及诱导生成小鳞茎时,使用较多的培养基组合为MS+6-BA(0.5~10.0)mg/L+NAA(0.01~3.0)mg/L或MS+6-BA(0.5~10.0)mg/L+2,4-D(0.1~2.0)mg/L;(3)常用的生根培养基为1/2MS+NAA(0.03,0.05,0.1,0.2,0.5)mg/L或1/2MS+NAA(0.03,0.05,0.1,0.2,0.5)mg/L+IBA(0.03,0.1,0.2)mg/L;(4)无论是多花水仙(Narcissus tazetta L.)还是黄水仙(Narcissus pseudonarcissus L.)的组织培养与植株再生的研究,在培养方案体系上都有较高的一致性.本文的研究对于进一步开展水仙的组织培养研究具有参考意义.The typical research results of tissue culture and plant regeneration of Narcissus in the past 40 years at our country and abroad are summarized in this paper.Analyzing these investigation results,some common protocols are got:The first,the most convenient and efficient in vitro tissue culture and plant regeneration material of Narcissus is"twin scale with bulb disk"or"twin scale";The second,when using scales with bulb disc or simple scales to induce callus and small bulbs,the relatively fixed plant hormone matching mode is MS+6-BA(0.5~10.0)mg/L+NAA(0.01~3.0 mg/L)or MS+6-BA(0.5~10.0)mg/L+2,4-D(0.1~2.0)mg/L;The third,the most commonly used medium for inclucing of roots is 1/2MS+NAA(0.03,0.05,0.1,0.2,0.5)mg/L or 1/2MS+NAA(0.03,0.05,0.1,0.2,0.5)mg/L+IBA(0.03,0.1,0.2)mg/L;Fourth,it is found that tissue culture and plant regeneration of both Narcissus tazetta L.and Narcissus pseudonarcissus L.accord with the above characteristics in the culture technique system.The characteristics of these technologies are of reference significance for us to carry out the further tissue culture research of Narcissus.
