益气活血方及其拆方对出血性血小板病患者静脉血iRhom2及GPⅤ表达的影响

摘　　要：目的观察益气活血方及其拆方对出血性血小板病患者静脉血非活性的菱形蛋白(inactive rhomboid-like protein,iRhom)2及血小板膜糖蛋白(platelet membrane glycoprotein,GP)Ⅴ表达的影响。方法选取33例出血性血小板病患者及30例血小板聚集检测无异常发现的健康献血员的静脉血,体外加益气活血方及其拆方进行中药孵育后,采用Real-time PCR法检测iRhom2 mRNA的相对表达水平,采用ELISA法检测GPⅤ蛋白表达水平。结果阴性对照组iRhom2 mRNA相对表达水平明显高于正常对照组,GPⅤ蛋白表达水平明显低于正常对照组(P<0.05)。全方组、益气活血组iRhom2 mRNA相对表达水平明显低于阴性对照组,GPⅤ蛋白表达水平明显高于阴性对照组(P<0.05)。结论益气活血方全方及其部分拆方治疗出血性血小板病的机制可能与下调iRhom2 mRNA表达及上调GPⅤ蛋白表达有关。Objective To observe the effects of Yiqi Huoxue decoction and its disassembled formulas on the expression of inactive rhombic like protein(iRhom)2 and platelet membrane glycoprotein(GP)V protein in venous blood of patients with hemorrhagic thrombocytopathy.Methods The venous blood of 33 patients with hemorrhagic thrombocytic and 30 healthy blood donors who were detected without abnormalities in platelet aggregation test were selected.The venous blood samples were supplemented and incubated with Yiqi Huoxue decoction and its disassembled formulas in vitro.The relative expression levels of iRhom2 mRNA were determined by the Real-time PCR assay,and the expression levels of GPⅤprotein were detected by ELISA.Results The relative expression levels of iRhom2 mRNA in the negative control group were significantly higher than those in the normal control group,and the expression levels of GP V protein were significantly lower than those in the normal control group(P<0.05).The relative expression levels of iRhom2 mRNA in the whole formula group,the Yiqi Huoxue group were significantly lower than those in the negative control group,and the expression levels of GP V protein were significantly higher than those in the negative control group(P<0.05).Conclusion The mechanism of Yiqi Huoxue decoction and its disassembled formulas in the treatment of hemorrhagic thrombocytic might be related to the downregulation of iRhom2 mRNA expression and upregulation of GP V protein expression.
