摘 要:目的鉴定米邦塔仙人掌的化学成分并进行定量分析。方法采用高效液相色谱法(HPLC)和核磁共振波谱分离和鉴定化合物。建立了同时测定两种成分的定量方法:采用Kromasil100AC1 8色谱柱(250 mm×4.6 mm,5μm),以乙腈(A)-0.1%甲酸水(B)溶液为流动相,梯度洗脱,检测波长为275 nm,流速1.0 ml/min,柱温30℃;进样量10μl。结果从米邦塔仙人掌中分离鉴定出2个化合物,分别是5-羟甲基-2-呋喃甲醛(1)和番石榴酸(2),番石榴酸为首次在该植物中分离得到。定量方法的精密度、重复性和稳定性均符合要求,5-羟甲基-2-呋喃甲醛和番石榴酸线性范围分别为5~150μg/ml(R^(2)=0.9992),10~1200μg/ml(R^(2)=0.9997),加样回收率均在90%~110%范围内,RSD均小于3%。结论本文建立了一种准确稳定地检测米邦塔仙人掌中化学成分的方法,可为其开发利用提供参考。Objective To identify and analyze quantitatively the chemical composition of Opuntia milpa alta. Methods HPLC and NMR were used to separate and identify the compounds, and a quantitative method was established for the simultaneous determination of the two components. For the determination, Kromasil 100 A C18 chromatographic column(250 mm×4.6 mm, 5 μm) was adopted, using acetonitrile(A)-0.1 % formic acid(B) aqueous solution as mobile phase for gradient elution. The detection wavelength was set at 275 nm, the flow rate was 1.0 ml/min, the column temperature was 30 ℃, and the injection volume was 10 μl. Results Two compounds were isolated from Opuntia milpa alta, which were 5-hydroxymethyl-2-furanformaldehyde(1) and piscidic acid(2). Piscidic acid was isolated from the plant for the first time. The precision, repeatability and stability of the quantitative method met the requirements. The linear ranges of 5-hydroxymethyl-2-furazaldehyde and piscidic acid were 5-150 μg/ml(R^(2)=0.9992)and 10-1200 μg/ml(R^(2)=0.9997), respectively. The recoveries were within 90 %-110 %, and all RSDs are less than 3 %. Conclusion In this experiment, an accurate and stable method is established for determination of the chemical constituents in Opuntia milpa alta.
