5/6肾脏切除动物疾病模型的中医证候探索及扶正化瘀降浊通络方证测验

摘　　要：目的:探讨5/6肾切除大鼠的中医证型及用扶正化瘀降浊通络方进行方证测验。方法:将雄性SD大鼠随机分为假手术组、模型组、中药组(给予扶正化瘀降浊通络方),采用“5/6肾切除法”造模,动态观察一般情况,给药后4、8、12周通过检测免疫指标CD4^(^(+))、CD8^(^(+))细胞比率评估“正虚”情况,检测微炎症指标C反应蛋白(c reactive protein,CRP)、肿瘤坏死因子-α(Tumor Necrosis Factor-α,TNF-α)、白介素-1β(Interleukin-1β,IL-1β)、白介素-6(Interleukin-6,IL-6)水平评估“邪实”情况。结果:模型组与假手术组比较,大鼠出现精神萎靡、毛发黄、目耳淡等本虚表现,伴舌紫黯、四肢肿胀等标实表现,全血中CD4^(^(+))降低、CD8^(^(+))升高,血清CRP、TNF-α、IL-1β、IL-6等微炎症因子表达增加(P<0.05,P<0.01);中药组与模型组比较,大鼠“本虚标实”的宏观表现有所改善,CD4^(^(+))升高、CD8^(^(+))降低,CRP、TNF-α、IL-1β、IL-6等微炎症因子表达量降低(P<0.05,P<0.01)。结论:5/6肾切除模型有“正虚邪实证”的宏观及微观表现,可作为“正虚邪实证”的动物证候模型,扶正化瘀降浊通络方可改善5/6肾切除大鼠“正虚邪实证”。Objective:To explore the traditional Chinese medicine(TCM)syndrome of Chronic Renal Failure(CRF)classic animal models,called 5/6 nephrectomy model and to test the syndrome with Fuzheng Huayu Tongluo prescription.Methods:Male SD rats were randomly divided into sham operation group,model group and TCM group which was administrated with Fuzheng Huayu Jiangzhuo Tongluo prescription.The model was established by removing 5/6 of the kidney.The general conditions of rats were observed dynamically.The rats were sacrificed in batches 4,8 and 12 weeks after administration.The blood was detected for the ratio of CD4+T cells and CD8+T cells as immune indicators to assess the condition of"Healthy Qi Deficiency".Serum micro-inflammatory markers,such as CRP,TNF-α,IL-1,and IL-6 were measured to assess the condition of"Evil Qi Excess".Results:Compared with the sham operation group,the rats in the model group showed symptoms of deficiency syndrome,such as listlessness,yellow and mottled fur,pale eyes and ears,etc.together with purple tongue and swollen limbs.The CD4+in the whole blood was decreased,the CD8+was increased.And the expression of microinflammatory factors,such as CRP,TNF-α,IL-1β,IL-6 and were increased(P<0.05,P<0.01).Compared with the model group,the symptoms of rats in the TCM group improved.CD4+increased and CD8+decreased,and the expression of CRP,TNF-α,IL-1β,IL-6 decreased(P<0.05,P<0.01).Conclusion:Rats from model group showed the macro and micro manifestations of"Healthy Qi Deficiency and Evil Qi Excess",which can be used as an animal syndrome model for"Healthy Qi deficiency and evil Qi excess".Fuzheng Huayu Jiangzhuo Tongluo prescription can improve the condition of"Healthy Qi deficiency and evil Qi excess"in rats treated with 5/6 nephrectomy.
