摘  要:为阐明一个大约克猪群体的遗传多样性,本实验用Chelex-100方法从来自四川省凉山州的30头大约克猪乳中提取基因组DNA,并用PCR方法扩增mtDNA D-loop区部分序列.结果显示,通过优化PCR循环数等参数,可以扩增出乳源基因组DNA的mtDNA D-loop区,产物可直接测序,获得364 bp的序列.30个大约克猪mtDNA D-loop区单倍型多样度(Hd)为0.618;核苷酸多样度(Pi)为0.00860,显示了较丰富的遗传多样性;检测到11个多态位点,界定了7种单倍型,其中具有Hap3单倍型的个体比例占50%以上.结果表明,乳源基因组DNA可用于mtDNA D-loop区的PCR扩增及直接测序,试验大约克猪母系遗传多样性较高.In order to assay the genetic diversity of a Yorkshire pig population,genomic DNA was extracted from 30 milk samples of Yorkshire pigs using Chelex-100 method,and partial mtDNA D-loop region was amplified by PCR and sequenced.The results showed that the mtDNA D-loop region could be amplified successfully using milk derived genomic DNA,and the PCR products could be directly sequenced,resulting in a 364 bp sequence.The haplotype diversity(Hd)and nucleotide diversity(Pi)of 30 Yorkshire pigs mtDNA D-loop were 0.618 and 0.00860,respectively,indicating a relevant high genetic diversity of this population.Eleven polymorphic sites,defining 7 haplotypes were detected,and more than 50%of the individuals carried Hap3 haplotype.The present results suggested that milk derived genomic DNA could be used for PCR amplification of the mtDNA D-loop region and subsequent direct sequencing,and the maternal genetic diversity of the experimental Yorkshire pigswas relatively abundant.
