摘  要:目的:探究舒郁胶囊在大鼠大脑组织中的化学成分分布及对海马神经元原代细胞5-羟色氨3受体(5-HT;R)功能的影响。方法:正常大鼠灌胃给予舒郁胶囊内容物后,取大鼠海马、额叶、纹状体、下丘脑、小脑和脑干组织,采用HPLC-ESI-MS;法检测上述大脑核团中的化学成分;复制大鼠抑郁情绪模型,灌胃舒郁胶囊内容物并提取大鼠含药血清,利用Western blotting法检测经各组灭活血清干预后的海马神经元原代细胞5-HT;R蛋白表达,利用全细胞膜片钳技术记录经各组灭活血清干预后的5-HT;R介导的通道离子电流。结果:在大鼠海马和额叶核团检测到5个化学成分,其中2个为新代谢产物;与正常组血清所孵育的海马神经元比较,抑郁组大鼠5-HT;R蛋白表达量及5-HT;R电流密度值均明显升高(P<0.05);与抑郁组血清所孵育的海马神经元比较,舒郁组大鼠5-HT;R蛋白表达量及5-HT;R电流密度值均明显降低(P<0.05)。结论:芍药苷、芍药内酯苷、芍药苷甲酯、去羟基去亚甲基芍药苷和柴胡皂苷元脱氧化物可能是舒郁胶囊调节5-HT;R通道功能发挥抗抑郁作用的主要化学成分。Objective:To analyze the chemical distribution of Shuyu capsule in rat brain tissue and its effect on the function of 5-HT;R in hippocampal neurons.Methods:The hippocampus,frontal lobes,striatum,hypothalamus,cerebellum and brainstem were collected respectively after intragastric administration of Shuyu capsule,and the chemical components in above brain tissue were analyzed by HPLC-ESI-MSn.The depression model of rats was successfully replicated,the contents of Shuyu capsule were perfused and the drug containing serum was extracted.The 5-HT;R protein expression levels in primary cells of hippocampal neurons were detected by Western blotting(WB)after the intervention of inactivated serum.And the 5-HT;R channel current was recorded by the whole-cell patch clamp.Results:A total of 5 compounds including 2 new metabolites were identified in the hippocampus and frontal lobes.Compared with the hippocampal neurons incubated by serum of normal group,the 5-HT;R protein expression and the current density value of 5-HT;R in depressive model group were significantly increased(P<0.05).Compared with the hippocampal neurons incubated in the serum of the depressive model group,the expression of 5-HT;R protein and the value of 5-HT;R current density in Shuyu capsule group were significantly decreased(P<0.05).Conclusion:Paeoniflorin,albiflorin,methyl paeoniflorin,dehydroxy methylene paeoniflorin,and saikosaponin deoxidation may be the main chemical component of Shuyu capsule to regulate the function of 5-HT;R channel and exert the antidepressant effect.
