摘 要:目的:探究玉竹多糖对油酸诱导的HepG2脂肪堆积的改善作用及其作用机制。方法:将实验分成正常对照组、模型对照组、洛伐他汀组、玉竹多糖低剂量组和玉竹多糖高剂量组,给药2 h后使用0.25 mmol/L油酸孵育24 h诱导产生脂肪堆积。油红O染色法检测细胞内脂滴形成,生化试剂盒法检测各组细胞总甘油三酯(TG)和总胆固醇(TC)含量,以及超氧化物歧化酶(SOD)和活性氧(ROS)水平。Western blotting法检测各组NF-κB/TLR4蛋白表达水平。结果:50μg/mL玉竹多糖能明显降低油酸诱导的HepG2脂滴堆积,降低模型细胞的TC/TG含量(P<0.05),降低模型细胞ROS水平并提高SOD活性(P<0.05),上调模型细胞NF-κB/TLR4蛋白表达水平(P<0.05)。结论:玉竹多糖能改善油酸诱导的HepG2脂肪堆积,其作用机制可能是基于抑制NF-κB信号通路产生的氧化应激损伤保护作用。Objective:To investigate the ameliorative effect and it’s mechanism of polygonatum odoratum polysaccharide(POP)on lipid accumulation induced by oleic acid in HepG2 cells.Methods:The experiment was divided into normal control group,model control group,lovastatin group,low dose of POP group,and high dose of POP group.After drug administration for 2 hours,fat accumulation was induced by incubation with0.25 mmol/L oleic acid for 24 hours.The formation of lipid droplets in cells was detected by oil red O staining,and the contents of total triglyceride(TG)and total cholesterol(TC),superoxide dismutase(SOD)and reactive oxygen species(ROS)were detected by biochemical kit.The expression of NF-κB/TLR4 Protein for each group were detected by Western blotting.Results:50μg/mL POP could significantly reduce the lipid accumulation in HepG2 cells induced by oleic acid,significantly reduce the content of TC/TG in model cells(P<0.05),reduce the level of ROS and increase the activity of SOD in model cells(P<0.05),and up regulate the expression level of NF-κB/TLR4 in model cells(P<0.05).Conclusion:Polygonatum odoratum polysaccharide can improve fat accumulation in HepG2 cells induced by oleic acid,and its mechanism may be based on the protective effect of inhibiting oxidative stress induced by NF-κB signaling pathway.
