摘  要:目的探讨ABCG2基因rs2231142位点基因多态性与痛风易感性的关系。方法选择293例男性痛风患者纳入痛风组,500例男性健康体检者纳入对照组。观察两组ABCG2基因rs2231142位点基因多态性。分析痛风发病的危险因素,不同遗传模型与痛风易感性的关系,以及基因与环境的相乘交互作用和相加交互作用。结果痛风组ABCG2基因rs2231142位点GT、TT基因型频率及等位基因T频率高于对照组,GG基因型、等位基因G频率低于对照组(P均<0.05)。高尿酸血症、高血糖和高密度脂蛋白是痛风发病的危险因素(P<0.05,OR>1.00),规律运动是痛风发病的保护因素(P<0.05,OR<1.00)。不同遗传模型中,TT基因型均表现为痛风的危险因素(加性模型OR=3.36,95%CI 2.18~5.19;显性模型OR=3.50,95%CI 2.55~4.81;隐性模型OR=5.18,95%CI 3.45~7.78)。ABCG2基因rs2231142位点GT+TT基因型与规律运动在相乘和相加模型中均存在交互作用,表现为痛风发病的保护因素(P<0.05,OR<1.00);TT基因型与规律运动仅在相加模型中表现出交互作用,表现为痛风发病的危险因素(P<0.05,OR>1.00)。结论ABCG2基因rs2231142位点基因多态性与痛风易感性间存在关联,等位基因T为痛风发作的危险因素且与规律运动间存在交互作用。Objective To explore the relationship between the polymorphism of rs2231142 loci of ABCG2 gene and susceptibility to gout.Methods We selected 293 male gout patients(gout group)and 500 male healthy controls(control group)as the research objects.We observed the polymorphism of rs2231142 loci of ABCG2 gene in the two groups.Then,we analyzed the risk factors of gout,the relationship between different genetic models and susceptibility to gout,and multiplicative and additive interactions between genes and environment.Results The frequencies of GT and TT genotypes and T allele for rs2231142 loci of ABCG2 gene were higher in the gout group than in the control group,but the frequencies of GG genotype and G allele were lower than those of the control group(all P<0.05).Hyperuricemia,hyperglycemia and high-density lipoprotein were risk factors for gout(all P<0.05,OR>1.00)and regular exercise was the protective factor for gout(P<0.05,OR<1.00).TT genotype was risk factor for gout in different genetic models(additive model:OR=3.36,95%CI:2.18-5.19,dominant model:OR=3.50,95%CI:2.55-4.81,and recessive model:OR=5.18,95%CI:3.45-7.78).The GT+TT genotype of rs2231142 loci with regular exercise was the protective factor for gout both in multiplicative and additive interactions(P<0.05,OR<1.00),but TT genotype with regular exercise was the risk factor for gout only in additive interaction(P<0.05,OR>1.00).Conclusion There is association between rs2231142 loci of ABCG2 gene polymorphism and gout susceptibility,and T allele is the risk factor for gout and has interaction with regular exercise.
