摘  要:丝裂原相关蛋白激酶(MAPK)信号通路在血吸虫的发育和产卵过程中发挥重要的调节作用,已有研究表明p38MAPK可调控曼氏血吸虫的发育,并参与雌性成熟过程。为此,本研究对日本血吸虫p38MAPK(Sjp38MAPK)进行了克隆、表达,并对其功能进行了初步分析。结果表明:Sjp38MAPK长度1038 bp,预测分子量39.58 kDa;生物信息学分析表明Sjp38MAPK与曼氏血吸虫和埃及血吸虫的同源性分别为93%和90%;qRT-PCR分析表明Sjp38MAPK在成虫中的表达量较高于虫卵、尾蚴和童虫时期;进一步利用siRNA抑制Sjp38MAPK表达,发现虫体卵壳蛋白的合成,运动以及细胞活性等方面受到影响。总之,本研究初步表明了Sjp38MAPK在日本血吸虫的发育和产卵过程中可能同样发挥重要的调节作用,值得深入研究。Mitogen associated protein kinase (MAPK) signaling pathway plays an important regulatory role in the regulation of development and sexual maturation in many organisms.Previous studies have indicated the important regulatory role of p38MAPK in development of Schistosoma mansoni.In the present study,we characterized S.japonicum p38MAPK (Sjp38MAPK) and evaluated its role in worm motility and cell viability.Bioinformatic analysis revealed that Sjp38MAPK shared 93% and 90% sequence identities with S.mansoni and S.haematobium,respectively.Phylogenetic tree analysis showed evolutionary closeness to other schistosoma spp.qRT-PCR analysis indicated the relatively high expression in adults at the transcriptional levels as compared with that in eggs,cercariae and schistosomula.siRNA silencing of Sjp38MAPK could affect the expressions of eggshell protein genes and influence worm motility and cellular viability.Overall,our preliminary study suggested the important role of Sjp38MAPK in development and egg production inS.japonicum.
