摘 要:1型鸭疫里默氏杆菌在华南地区流行,本试验在获得1型鸭疫里默氏杆菌天然弱毒株SG株的基础上,采用具有在线控制pH和溶解氧(DO)的5 L发酵罐,通过对发酵培养基、发酵培养过程中DO值、发酵培养转速进行优化,获得1型鸭疫里默氏杆菌弱毒株SG株。结果显示:SG株发酵培养的最适培养基配方中每升培养基含胰蛋白胨17.5 g、大豆蛋白胨7.5 g、酵母提取物7.5 g、葡萄糖2.5 g;SG株发酵培养的最适DO值为20%、最适转速为150 rpm,当发酵8 h活菌数达到高峰(1.27×10^(10)CFU/mL);以获得的优化工艺在GMP 生产车间规模化发酵生产 3 批鸭疫里默杆菌弱毒株SG株,收获弱毒菌液的活菌数均高于1.0×10^(10)CFU/mL。本试验为鸭疫里默氏杆菌弱毒疫苗抗原的高效制备提供了指导。Riemerella anatipestis type 1 is prevalent in South China.In this experiment,industrial propagation of the attenuated SG strain of R.anatipestis type 1 was developed and optimized using 5 L fermentation tank with on-line control of pH and dissolved oxygen (DO) regarding the liquid medium,DO value and fermentation speed in the fermentation process.The optimum fermentation medium per liter for SG strain contained 17.5 g tryptone,7.5 g soybean peptone,7.5 g yeast extract and 2.5 g glucose.The optimum DO value and rotational speed of SG strain fermentation were 20% and 150 rpm,respectively.The number of viable bacteria reached a peak(1.27 ×10^(10)CFU/mL) after 8 hours of fermentation.Three batches of the attenuated SG strain were produced in a GMP workshop for large-scale industrial fermentation and the viable bacteria of each batch was higher than 1.0×10^(10)CFU/mL at harvest.This experiment provided preliminary guidance for the efficient preparation of the attenuated vaccine antigen of R.anatipestis.
