摘 要:目的:探讨针刺“百会”“大椎”穴对短暂性全脑缺血再灌注大鼠脑组织自噬相关蛋白的影响及作用机制。方法:将54只雄性SD大鼠(200~300 g)随机分为模型组、非穴位组及针刺组各18只。应用改良三血管闭塞法制备短暂性全脑缺血再灌注(transient global cerebral ischemia reperfusion,TGCIR)大鼠模型,针刺组及非穴位组分别针刺百会穴、大椎穴及两穴平行线旁开1 cm非穴位处,每天2次,共3天,治疗3天对大鼠进行神经功能缺损评分,检测大鼠患侧海马CA1(hippocampal,CA1)区脑组织自噬相关蛋白微管连接蛋白轻链3(microtubule-associated protein light chain 3,LC3)及苄氯素1(Beclin 1)表达。结果:与模型组比较,非穴位组在神经功能评分及自噬相关蛋白表达方面均无明显差异(P>0.05);与模型组、非穴位组比较,针刺百会、大椎穴能改善大鼠神经功能评分(P<0.05),上调LC3、Beclin 1蛋白表达(P<0.05)。结论:针刺百会、大椎穴能够上调短暂性全脑缺血再灌注大鼠自噬相关蛋白LC3、Beclin 1表达,进而改善脑损伤大鼠神经功能缺损。Objective:To explore the influence of acupuncture at Baihui(DU20)and Dazhui(BU14)on the autophagy related protein of cerebral tissue in rat models with transient global cerebral ischemia reperfusion(TGCIR),and its mechanism.Methods:All 54 male SD rats(200 to 300g)were randomized into the model group,non acupoint group and acupuncture group,18 rats in each group.Rat models with TGCIR were prepared using modified three vessel occlusion method,non acupoints group and acupuncture group were given acupuncture(Baihui and Dazhui)and non 1 cm beside the parallel line of the two points twice each day,for three days to perform NIHSS after 72 hours of intervention,to detect the expressions of the autophagy related protein of cerebral tissue LC3 and Beclin1 in the CA1 area of affected side in rats.Results:Compared with the model group,non acupoint group showed no obvious difference in NIHSS and the expressions of the autophagy related protein(P>0.05).Compared with the model group and non acupoint group,acupuncture at Baihui and Dazhui could improve NIHSS(average P<0.05),up regulate the expressions of LC3 and Beclin 1 protein(P<0.05).Conclusion:Acupuncture at Baihui and Dazhui could up regulate the expressions of the autophagy related protein LC3 and Beclin1,therefore to improve neurological deficit in rats with cerebral injury.
