摘  要:将对数生长期的布朗葡萄藻Botryococcus braunii接种到250 m L三角瓶培养体系中,无菌培养液体积为150 m L,分别选择富营养的BG-11培养基、贫营养的f/2培养基及营养水平中等的改良Chu培养基和SE培养基,初始OD690均控制在0.02左右。光暗比12 h∶12 h、光照强度4 000 lx、(25±2)℃下一次性摇床培养18 d,转速100 r/min,比较了布朗葡萄藻在不同营养供给条件下的生长速度、物质积累和单位产率。结果发现:布朗葡萄藻生长速度取决于培养液的营养元素贫富水平,富营养的BG-11培养基中的比生长速率高于中营养的Chu和SE培养基,而贫营养的f/2培养基中比生长速率最低。但是,富营养的BG-11不利于细胞总脂和总烃的积累,而贫营养f/2培养基更利于布朗葡萄藻细胞积累总脂、总烃和类胡萝卜素。布朗葡萄藻的单位物质产率取决于生长速率和单个细胞物质积累的综合效应;相对而言,贫营养培养基f/2更适合于布朗葡萄藻生产与积累储存性物质(特别是油脂)。所以,富营养的培养基,如BG-11,适合于布朗葡萄藻的快速生长;贫营养的培养基,如f/2,适合于油脂生产与积累;而从营养成分的投入和生产物质的产出效率看,f/2培养基显著优于其他三种培养基。Alga Botryococcus braunii in the logarithmic growth phase was inoculated into a 250 mL Erlenmeyer flask with a sterile culture medium volume of 150 mL. In the culture of the alga, nutrient-rich BG-11 medium, poor nutrient f/2 medium, the modified Chu medium, and SE medium with medium nutrient level were used, respectively. The alga was cultured with the initial OD;of about0.02, at light∶dark=12 h∶12 h with light intensity of 4 000 lx, and at(25±2)℃ for 18 d on a shaker of 100 r/min.The growth rate,material accumulation and yield of the alga under different nutrient supply conditions were compared. It was found that the growth rate of the alga depended on the richness of nutrient elements in the medium, with higher specific growth rates in the highest nutrient-rich medium of BG-11 than those in the middle nutrient-rich media of Chu medium and SE medium, and the poorest growth in the lowest nutrient-rich medium of f/2. However, the f/2 medium was shown to be more beneficial for the alga to accumulate lipids, hydrocarbons and carotenoids, while the highest nutrient-rich medium of BG-11 was not. The materials yields of the alga were dependent on the comprehensive effect of growth rate and material accumulation per cell. In comparison, the lowest nutrient-rich medium of f/2 was more suitable for the production and accumulation of stored substances(especially lipids)in the alga. Therefore, the rich nutrient media like BG-11 was the suitable medium for the growth while the poor nutrient media like f/2 was the suitable medium for lipid production by the alga. In addition, the f/2 medium was significantly more efficient than the other three in terms of input-output ratio of nutrient utilization.
