增食欲素A调节大鼠肾上腺嗜铬细胞瘤PC12细胞自噬的研究

摘　　要：目的探讨增食欲素A(orexin-A)对大鼠肾上腺嗜铬细胞瘤细胞自噬的影响。方法分别用10^(-6) mol/L orexin-A和10^(-6) mol/L orexin-A+增食欲素受体1(OX_(1)R)抑制剂作用于大鼠肾上腺嗜铬细胞瘤PC12细胞,另设1个空白对照组。通过Western blotting和ELISA方法测定PC12细胞中OX_(1)R、LC3Ⅱ/LC3Ⅰ、Beclin-1蛋白的表达水平和去甲肾上腺素(NE)的分泌水平,并在电镜下观察空白对照组和orexin-A组PC12细胞的自噬情况。结果电镜下可见orexin-A组PC12细胞的自噬体较空白对照组增多。orexin-A组OX_(1)R、LC3Ⅱ/LC3Ⅰ和Beclin-1蛋白的表达水平和NE的分泌量均显著高于空白对照组和orexin-A+OX_(1)R抑制剂组,差异有统计学意义(P<0.05)。结论orexin-A可通过OX_(1)R促进PC12细胞自噬,并能促进PC12细胞分泌NE。Objective To determine the effects of orexin-A on autophagy of rat adrenal pheochromocytoma(PC12)cells.Methods PC12 cells were treated with orexin-A(10^(-6) mol/L)and orexin-A(10^(-6) mol/L)with orexin receptor-1(OX_(1)R)inhibitor.A blank control group was also included.OX_(1)R,LC3Ⅱ/LC3Ⅰ,and Beclin-1 protein expresssion was determined by Western blotting.Norepinephrine(NE)secretion volume was measured by enzyme-linked immunosorbent assay(ELISA).Autophagy of PC12 cells in the blank group and the orexin-A group was observed using an electron microscope.Results PC12 cells treated with orexin-A showed higher autophagy than control cells.Expression of OX_(1)R,LC3Ⅱ/LC3Ⅰ,and Beclin-1 and NE secretion were significantly higher in the treated cells than in the blank control group and the orexin-A+OX_(1)R inhibitor group(P<0.05).Conclusion Orexin-A can promote autophagy of PC12 cells through OX_(1)R and promote secretion of NE in PC12 cells.
